電解氧化水與振動方式對於接菌雞蛋之殺菌效果

Efficacy of Electrolyzed Oxidizing Water Combining with Agitation in Sanitization of Spiked Shell Egg

摘要


本研究探討不同殺菌液對純菌及殺菌液配合振動方式對接菌雞蛋之殺菌效果。沙門氏桿菌(Salmonella spp.)、大腸桿菌(Escherichia coli)及金黃色葡萄球菌(Staphylococcus aureus)菌液(9.29 log CFU/ml)分別以含氯(total available chlorine)約20 ppm的EO water(EOWM)振動30 sec,所有接種菌被完全殺滅。Salmonella菌液經過EOWL振動30 sec,降低菌數比以NaOCl溶液(含氯20 ppm)多降約1.8 log CFU/ml,振動較長則降低菌數較多。含氯約10 ppm的EO water(EOWL)對於三種菌的殺滅效果依序為E. coli、S. aureus及Salmonella。EOWH或NaOCl溶液配合靜置10 min對於蛋殼上Salmonella之降低數均比EOWM多且有顯著差異(p<0.05)。三種殺菌液振盪30 sec或超音波30 sec對Salmonella均有明顯效果。EOWH以三種振動方式對於E. coli降菌數多且差異顯著,NaOCI以靜置或振盪則對E. coli殺菌效果同EOWH,但NaOCI以超音波之降低效果則比EOWH者較差,又EOWM以靜置或振盪對於E. coli降低效果均比EOWH組較差。EOWH以三種振動對於蛋上的S. aureus降低數比其它殺菌液者多且差異顯著,EOWM及NaOCl溶液以三種振動對S. aureus降低數效果相似約5 log CFU/egg。各檢液以胰化酪蛋白大豆培養液35℃培養24 hr之澄清管數,可快速判定各處理組之殺菌效果。

The aim of this study was to investigate the effects of chlorinated sanitizers combining with agitation on the bacterial reduction in spiked shell egg. Electrolyzed oxidizing water (EOW) with chlorine concentration at 20 ppm (EOWM) was found effective when pure cultures of Salmonella spp., Escherichia coli and Staphylococcus aureus was used. EOWM was not as effective as EOW with a higher chlorine concentration at 40 ppm (EOWH) when spiked eggs were tested. Agitation with gentle shaking or ultrasonic were found most effective when used in combination with EOWM for eggs spiked with the above three types of bacteria, resulting in a 5 to 6 logs of reduction in bacterial population. Comparable results were obtained with sodium hypochorite (NaOCl) solution with a chlorine concentration at 20 ppm and were used in combination with agitation. Agitation appeared to promote the bactericidal effects of both EOW and NaOCl solution. A rapid test, based on growth in trypticase soy broth (TSB), to monitor the bactericidal efficacy of sanitization treatment was developed. Two eggs after treatment were washed with 25 ml of peptone buffer, and 0.1 ml aliquot was used to inoculate four tubes each containing 10 ml of TSB and incubated at 37°C for 24 hours. When all 4 tubes showed negative growth, the sanitization treatment was judged to be successful in which at least a 5 to 6 logs of reduction in bacterial population was achieved.
