近年來益生菌代謝物-胞外多醣體已經引起廣泛興趣與討論,但對其相關功能瞭解甚少,本研究主要探討新鮮水果培養基培養益生菌對益生菌生長狀況與胞外多醣體 生成以及對巨噬細胞J774A.1生長的影響。本研究使用的水果為香蕉、蘋果、鳳梨,益生菌為標準菌Bifidobacterium longum BCRC 14634,及由泡菜中篩選出五碳醣片球菌/Pediococcus pentosaceus FY-001 (對照組),以離心及酒精萃取胞外多醣體,經0.22 μm濾膜過濾除菌後,以酚-硫酸法定量總醣量,做為外多醣生成量。在巨噬細胞J774A.1添加5 μg/ml胞外多醣體後,觀察細胞之増殖。以DPPH清除率測定抗氧化能力.結果顯示,三種水果培養基對益生菌增殖均優於MRS培養基,胞外多醣體生成量以香蕉培養基為最高,達4115.389 ± 405.88 mg/I,抗氧化活性則以鳳梨發酵物最高,為47.35±5.587%。胞外多醣體對巨噬細胞:F774A.1存活率的實驗顯示以香蕉與蘋果培養基最佳.本研究結果顯示,水果培基能替代MRS培養基,除了提供新式培養基,也證實水果培養基能促進益生菌増殖及增加胞外多醣體生成量,並促進巨噬細胞J774A.1存活率.本研究建議可以水果培養基培養之胞外多醣體做為一種免疫調節劑,改善腸胃道功能,並對抗腸胃道感染。
Exopolysaccharides (EPSs) are metabolites of probiotics which have gained wide interest recently. However, information regarding their function is limited. This study was aimed to investigate the growth and EPSs production of probiotics cultivated infresh fruits (banana, apple, and pineapple) media. Effects of EPSs on the growth of morophage J774A.1 were also investigated. Probiotics used in this study were Bifidobacterium Jongum BCRC 14634 and Pediococcus pentosaceus FY-001 screened from fermented vegetable as a control. EPSs separated by centrifugation and ethanol extraction and sterilized by 0.22 μm filter were detected by using phenol-sulfate method to measure the total carbohydrate. The proliferation of macrophages J774A.I was measured after treatment with 5 μg/ml EPS, and antioxidant activity was measured by DPPH scavenging effects . The results showed that the proliferations of B. Jongum BCRC 14634 in three kinds of fruit medium were higher than that in MRS medium. The content of EPS cultivated by banana medium was the highest (4115.39±405.88 mg/l ) . Pineapple EPS had the highest antioxidant activity, 47.35 ± 5.587%. As to the proliferation of macrophage J774A.l treated with EPS, banana and apple EPS were the better selection. Our research revealed that MRS medium can be replaced by fruit media. In conclusion, a new style of Lactobacillus media, we prove the evidence that fruit media could enhance the proliferation of probiotics, whose EPS could bean immunomodulator to improve the function of gut, against gastrointestinal infections
