西红花苷-1对实验性糖尿病小鼠肝保护作用研究

Protective Effect of Crocetin-1 on Liver in Experimental Diabetic Mice

摘要


观察西红花苷-1对链脲佐菌素(STZ)诱导的糖尿病小鼠肝保护作用。方法:采用STZ(180mg/kg)腹腔注射建立糖尿病小鼠模型。将造模成功后的小鼠随机分为3组(n=10):糖尿病模型(DM)组、西红花苷-1(C-1)组、阳性对照二甲双胍(MH)组。另取10只正常小鼠 设为正常对照(NC)组。给药组每天灌胃1次,连续6周,模型组和正常对照组灌胃生理盐水。给药第2、6周测定小鼠AST、ALT和ALP水平,给药第6周测定血清和肝脏中SOD、MDA、CAT、TNF-α和IL-6等指标的变化情况。与NC组相比,DM组血清AST、ALT、ALP、MDA和TNF-α水平均显著升高,SOD和MDA水平显著降低;肝脏TNF-α、MDA显著升高,SOD和CAT显著降低;与DM组相比,C-1组血清AST、ALT、ALP、MDA水平均显著降低,SOD和CAT水平显著均显著升高;肝脏TNF-α显著降低,CAT显著升高。西红花苷-1对链脲佐菌素诱导的糖尿病小鼠有一定肝保护作用,其机制可能和改善氧化应激和炎症有关,提示西红花苷-1可能用于糖尿病肝病的防治。

Observe the hepatoprotective effect of crocin-1 on streptozotocin (STZ)-induced diabetic mice. Diabetes mellitus mice were established by intraperitoneal injection of STZ (180 mg/kg). The 30 well-established diabetes mice were randomly divided into three groups: Diabetic mellitus (DM) group, Crocin-1(C-1) group and positive control (metformin hydrochloride MH) group. Another ten normal mice were selected as normal control (NC) group. The mice in C-1 and MH groups were intragastrically administered with C-1 60 mg/kg or MH 200mg/kg once a day for 6 weeks, mice in DM and NC were given normal saline. The levels of AST, ALT and ALP in mice were measured at the 2nd and 6th week after administration, and the changes of the indicators including SOD, MDA, CAT, TNF- and IL-6 in serum and liver were determined after 6 weeks of continuous administration. Compared with NC group, the levels of AST, ALT, ALP, MDA and TNF- in serum were significantly increased, while the levels of SOD and MDA were dramatically decreased in DM group. Compared with DM group, the levels of AST, ALT, ALP and MDA were significantly decreased with obviously increased levels of SOD and CAT in serum, and the TNF- of liver were starkly declined with a notable elevation of CAT in C-1 group. Crocin-1 has certain hepatoprotective effect on streptozotocin-induced diabetic mice, and its mechanism may be related to the improvement of oxidative stress and inflammation, which suggested that crocin-1 could be used for the prevention and treatment of diabetic liver disease.
