人工馴化培養蛹蟲草不同部位抗炎效果之研究

Study on the Anti-inflammatory Effects of Different Parts of Artificially Cultivated Cordyceps militaris

摘要


藥食兩用食材之常態食用,多為華人所喜愛的保健方式之一,其中人工馴化所培養之蛹蟲草(Cordyceps militaris)即是其一,近年來亦多有相關科學功效研究。然而,其培育之子實體與菌絲體(亦稱基座),其不同部位功效差異,則較少被研究,因此,本研究採用不同部位人工馴化所培養之蛹蟲草,研究素材區分有子實體與菌絲體,進行抗炎效果及其毒性分析之差異研究。研究使用經乾燥研磨至250微米粒徑,依三種不同濃度(0.25%、0.5%、1.0%)分別作用於巨噬細胞株(mouse BALB/c macrophage RAW 264.7),進行抗發炎活性三重複之檢測,包括一氧化氮(nitric oxide, NO)、介白素6(interleukin 6, IL-6)及腫瘤壞死因子-α(tumor necrosis factor-alpha, TNF-α)等三種細胞發炎激素,以及細胞毒性試驗(microculture tetrazolium assay, MTT assay)。結果顯示,對於受到細菌脂多醣(Lipopolysaccharide, LPS)活化的巨噬細胞株,觀察在250微米粒徑蛹蟲草子實體與菌絲體之水萃取液,在LPS所誘發NO、IL-6與TNF-α生成,具有抗發炎反應。結論:(1)添加子實體水萃取液1.0%,抑制率分別為93.9%、61.6%與46.8%,無細胞毒性反應。(2)添加菌絲體水萃取液1.0%,顯示抑制率分別為27.6%、48.5%及16.8%,無細胞毒性反應。總結,人工馴化培養蛹蟲草之子實體與菌絲體,除具顯著抗炎效果外,亦說明在產業應用上,菌絲體亦具備節省材料與成本之潛在效益。

The normal consumption of medicinal and edible ingredients is one of the favorite health care methods of the Chinese. Among them, the artificially cultivated Cordyceps militaris is one of them. In recent years, there have been many related scientific efficacy studies. However, the differences in efficacy of different parts of the cultivated fruiting bodies and mycelium have been less studied. Therefore, in this study, different parts of Cordyceps militaris were artificially domesticated and cultured, and the research materials were used to distinguish the fruiting bodies and mycelium to conduct a study on the differences in anti-inflammatory effects and toxicity analysis. The study used dried and ground to a particle size of 250 microns, and acted on the macrophage cell line (mouse BALB/c macrophage RAW 264.7) at three different concentrations (0.25%, 0.5% and 1.0%) to conduct anti-inflammatory activity in three repeat tests. The test includes three cell inflammatory hormones, including nitric oxide (NO), interleukin 6 (IL-6), and tumor necrosis factor-alpha (TNF-α), as well as cell Toxicity test (microculture tetrazolium assay, MTT assay).The results showed that for macrophage cell lines activated by bacterial lipopolysaccharide (LPS), it was observed that in the aqueous extract of Cordyceps militaris fruiting bodies and mycelium with a particle size of 250 μm, the NO, IL-6 and TNF-α induced by LPS were production, has anti-inflammatory response. Conclusion: (1) Adding 1.0% fruiting bodies water extract, the inhibition rates were 93.9%, 61.6% and 46.8% respectively, without cytotoxicity. (2) Adding 1.0% mycelium water extract showed inhibition rates of 27.6%, 48.5% and 16.8% respectively, with no cytotoxic reaction. In summary, the artificial domestication and cultivation of the fruiting bodies and mycelium of Cordyceps militaris not only has significant anti-inflammatory effects, but also shows that Cordyceps militaris also has the potential benefit of saving materials and costs in industrial applications.
