高黏著力之溫度敏感型水膠配方之開發及其藥物釋放研究

摘要


本實驗為水膠配方開發,藉由不同的配方組成、合成方法,來改變水膠的結構,進而影響其黏著力、熟成時間、膠體強度等物理性質。之後引入添加溫感型的高分子於水膠結構中,使其具有溫度敏感的特性。最後探討不同的水膠結構設計及物理性質,以及溫度敏感高分子複合水膠於藥物控制釋放系統之適用性。 本研究以丙烯酸(AA)、丙烯酸鈉(SA,sodium acrylate)之共聚物NP-700為主,並混以羧甲基纖維素鈉 (sodium carboxymethyl cellulose,CMC)構成水膠,並以乳化聚合方式合成之溫度敏感型poly(NIPAAm-co-AA) micro-gel水溶液引入水膠結構中,利用Al3+作為交聯劑,形成溫度敏感高分子複合水膠。觀察不同的水膠型態、熟成情形、交聯度對膨潤性質及黏著力的影響,並以熊果素為模擬藥物,進行複合水膠薄膜中藥物釋放與擴散性質研究,觀察摻混不同形態溫度敏感型乳液對複合水膠薄膜藥物控制釋放之影響。

In this research, different prescriptions and steps were adopted to change the structure of hydrogel. Thus, the adhesion force, time for mature and mechanical strength of hydrogel could be studied. Furthermore, by adding thermal responsive polymer to hydrogel, the complex hydrogel could be sensitive to temperature. Finally, composition and its physical properties of hydrogel, and the applicability of complex hydrogel in drug release control system was investigated. Acrylic acid / sodium acrylate copolymer (NP-700) and sodium carboxymethyl cellulose were used to syntheses hydrogel in the presence of aluminum ion to interact with carboxyl group in this research. Thermal responsive poly(NIPAAm-co-AA) micro-gel solution was obtained by emulsion polymerization and introduced to the hydrogel. The adhesion force, time for mature, crosslinking degree and swelling ratio of complex hydrogel were observed. Finally, Arbutin was the drug used in the experiment for researching the drug releasing and its diffusion mathematical model of complex hydrogel film. The effect of the morphology of the thermal sensitive polymer in complex hydrogel film on drug releasing was studied too.
