碳酸鈣磷酸鈣核殼載體之製備及鑑定
Preparation and Characterization of Core-Shell Carriers of Calcium Phosphate and Calcium Carbonate
摘要
碳酸鈣為自然界中常見的生物礦物之一，由於其無毒、高生物相容性與對 pH 值敏感等特質，是非常具有潛力的藥物載體。由於碳酸鈣在水中非常不穩定且無法分散，在本研究中我們嘗試於碳酸鈣的外層包覆磷酸鈣，利用具有更低水溶解度的磷酸鈣保護內層的碳酸鈣。本研究嘗試使用直接混合及氣體擴散法製備微米尺度的含鎂非晶相碳酸鈣顆粒，最後利用薄膜水合法成功製備，本研究也利用磷酸銫甲醯胺溶液成功合成出碳酸鈣磷酸鈣核殼微米顆粒，並嘗試將此核殼顆粒作為阿斯匹靈的載體，藥物大約佔總重量約 10%，此外，本研究類推使用此實驗概念以分散含鎂非晶相碳酸鈣奈米顆粒。

Calcium carbonate is one of the common biological minerals in nature. Because of its non-toxicity, high biocompatibility and good sensitivity to pH, it is a promising drug carrier. Since nano-particles of calcium carbonate is readily soluble in water, we proposed to coat calcium phosphate on the outer layer of calcium carbonate, and use calcium phosphate with lower water solubility to protect the inner layer of calcium carbonate and the drug incorporated inside. As a proof of concept, we took micron-sized Mg stabilized amorphous calcium carbonate (MgACC) as our model system. In this study, we have successfully synthesized calcium carbonate calcium phosphate core-shell microparticles using cesium phosphate formamide solution. In addition, the size and morphology of the core-shell MgACC were maintained after loading 10 wt % of aspirin. We also use this idea to disperse nano-sized MgACC.
