以生醫訊號模型初步探討多醣體對免疫細胞之影響

摘要


目前本研究對於癌症患者經由免疫治療方式所能達到預後效果,在此所投的藥物、時間、濃度和細胞的相對數量是很重要的。本論文提出一個能有效投藥的評估系統。這是藉由增強免疫效果方面的培養系統,以其所得的統計數據結果來設計出一個評估性平台所需要的相關參數。目前醫學上已知身體中NK細胞、T4細胞、T8細胞具有相當高的抗癌能力,因此我們利用多醣蛋白(β-Glucan)來加速這三種細胞的成長,並進行細胞分類上的比較,以觀察到底哪一種細胞所受到的影響為最大。 在實驗上,我們將受測者的血液檢體分成兩大組,分別是第一個人的細胞培養液和第二個人的細胞培養液,每一大組分成十二組被添加不同濃度的多醣蛋白,在二氧化碳培養箱中進行培養;並於不同天數進行顯微鏡下觀察與流式細胞分析儀分析,來看NK細胞、T4細胞及T8細胞的變化。我們一共做了三次試驗,以平均數值來分析。 我們利用SPSS來分析,首先以Levene’s test作變異數同質性檢定,以判斷是否為同一群組,再以one way ANOVA作統計學上的檢定,以觀測差異顯著性,最後用Tukey’s法作結果後分析,以進行成對比較,來觀測所得的實驗結果是否具有統計學上的意義。結果上發現,NK細胞於整體細胞中的增幅具有較為明顯的變化。因此可知多醣蛋白對NK細胞的生長刺激性質,在免疫治療上具有相當大的意義。

In immune therapy, it is very important to decide the dose and concentration ofβ-glucan. This research presents an analysis system to help serve this problem. In this system β-glucan is used to accelerate the growth of immune cells and compare the differences between NK cells, T4 cells, and T8 cells. The goal is to observe the most effected one. There are two subjects in this experiment. Twelve blood samples of each subject are given β-glucan of different concentrations. The samples were cultured in the CO2 incubator. The percentage change of cells was estimated via the flow cytometer. This experiment was repeated three times to eliminate any bias. SPSS was used to analyze the data. First, Levene’s test was used to examined the data. If it was significant, then one way ANOVA followed by Tukey’s test were used to process the comparison. We found that the percentage of NK cells increased significantly with β-glucan.
