舒緩類風濕性關節炎的保健食品功效評估

Evaluation of the healthy food with anti-rheumatoid arthritis effects

摘要


伴隨醫療科技日益進步及人民平均餘命延長,人口結構老化造成已成世界趨勢,退化性疾病成為亟需重視並應尋求預防的問題。越來越多研究指出有些慢性疾病可藉由生活習慣的改善而略為緩解。關節炎 (arthritis) 為人類疾病中最普遍的慢性關節疾病,而類風濕性關節炎是屬於自體免疫異常的一種關節炎,可能造成嚴重時的生理機能不正常。本研究在於探討建立預防或舒緩關節症狀保健功能之評估方法,研究方法包括蒐集現今可有效誘發關節炎的實驗動物模式及不同品系鼠種的誘發關節炎成效,找出適當的誘發關節炎的動物實驗模式,接著餵食本實驗室之前以體外實驗篩選出來具抗發炎潛力的食材苜蓿芽,進一步用以評估其他具免疫調節保健功效的食品。 本研究誘發關節炎的動物模式,是以致病機制及病程和人類的類風溼性關節炎致病模式較相似的膠原蛋白誘發關節炎(collagen induced arthritis,簡稱CIA)動物模式,免疫接種較具誘發成效的DBA/1J小鼠,再以苜宿芽餵食被誘發關節炎的小鼠,測相關生化與免疫指標。本研究以DBA/1J小鼠誘發類風濕性關節炎的成功率為85%,但評估經CIA的故以DBA/1J小鼠的CIA模式,於發炎高峰期(免疫接種七週)後分組管餵具抗發炎效果的苜蓿芽乙酸乙脂萃物 (ASEA) 測試,結果在藥物組亦未顯示有良好的舒緩類風濕性關節炎成效,經調整研究設計,於免疫接種的隔日即進行分組及管餵投予測試食材,餵食ASEA及市售免疫奶粉評估其抗發炎成效,結果顯示餵食ASEA及免疫奶粉,可降低小鼠發炎指標,包括:降低小鼠四肢的發炎腫脹指數;顯著降低血清中的抗體濃度,包括特異性抗體IgG2a濃度及發炎性抗體總 IgG濃度;降低Th1反應之發炎性細胞激素TNF-α、IFN-γ、IL-2及IL-1β的分泌量;降低調節Th1反應的IL-12 細胞激素分泌量。 綜合上述實驗結果可知,本研究以CIA模式免疫接種DBA/1J小鼠七週,並於免疫接種的一開始餵食保健食品的動物實驗模式,適合作為評估保健食材舒緩類風溼性關節炎的保健功效,且顯示本次研究的苜蓿芽乙酸乙酯萃取物及免疫奶粉在早期餵食的CIA誘發動物模式下,具有調節發炎性細胞指標T細胞,降低Th1反應,抑制促發炎細胞激素的分泌,降低發炎反應,進而達到舒緩類風濕性關節炎的功效。

Population aging is a world-wide tendency as for the great progress of medicine and extend of life expectancy. Degeneration disease should be gotten more attention and seek for solution and prevention urgently. There are more evidences show that chronic diseases can be alleviated by healthy diet pattern and life style. Arthritis is the common chronic disease in elderly. Rheumatoid arthritis which resulting from an overactive immune system is belonging to autoimmune disease and will cause serious disease. The purpose of this study is to focus on the anti-rheumatoid arthritic effect of functional food. In the study, we chose CIA (collagen induced arthritis) as animal model because it possesses many similarities to human RA. The CIA model immunization DBA/1J mice and then fed heathy food to evaluation the anti-rheumatoid arthritis effects. The efficiency of arthritis induction was 85% in CIA-DBA/1J mice. At first exam, the result didn’t show good anti-inflammation ability even in drug group. So we adjust feeding time immediate from the day after immunization. We found ASEA and hyperimmune milk show good anti-inflammation ability by decreasing total swelling score and decreasing the level of specific antibody IgG2a, inflammation antibody IgG and inflammation cytokine such as TNF-α、IFN-γ、IL-2 and IL-12. As a conclusion, we successfully evaluation of the healthy food with anti-rheumatoid arthritis effects by CIA_DBA/1J animal model and found that administration of ASEA or hyperimmune milk during the early stages of CIA development resulting in a modulating effect on inflammation markers and on the progress of symptoms of arthritis in animal, decrease Th1 reaction, inhibit secrete of inflammation cytokine and alleviate rheumatoid arthritis.
