抗原性醣類Globo H的合成

摘要


細胞表面上分布著許多碳水化合物的配位體,而這些碳水化合物的配位體已經過證實,有傳遞細胞與細胞間訊息的功能,像是細菌或者病毒入侵人體,也是透過此一機制,這也顯示出這些碳水化合物的重要性以及針對這些化合物做研究的興趣。近來有許多化學家對於醣類的合成有深入的研究,已經可以合成出複雜的多醣體,近一步將所合成出的多醣體拿來做多方面的應用,像是生物檢測,疫苗的研發,醣晶片的製作…等等。但是合成多醣體的步驟繁雜,又耗費許多時間,所以需要有更快速的方法來合成多醣體。這次研究是合成抗原性醣類glycosphingolipid Globo H,它大量分布在乳癌細胞表面,可能和乳癌細胞的增長以及惡化有極大的關聯。此醣分子也可用以製作醣晶片,以檢測病人抗體的存在。為了更快速的合成出Globo H,我這次研究使用programmable one-pot strategy來做醣類合成,當所有的building blocks都合成出來之後,直接用one-pot reaction來做成Globo H這個六醣體。而近半年來有研究指出,抗體會辨識的Globo H構造有好幾種,而兩個最大基本差異在於fucosyl group的有無,所以這次研究要設計適合的building blocks已用於未來合成Globo H類似物。

Abstract Cell surface are decorated with a myriad of glycoconjugates that mediate important cell-cell interactions. The invations of bacteria or virus onto human cells take place via a similar mechanism. Because of their biological significance, it is considered a critical challenge to efficiently synthesize complex carbohydrates, to provide enough quantity for biological investigation, such as vaccine development, carbohydrate microarray and drug discovery. This thesis research aims to synthesize the tumor-associated antigen that is highly expresses on the surface of breast cancer cells. In order to establish an expedition preparation, a new one-pot strategy was designed to form Globo H hexasaccaride or the analogue without a fucosyl group.
