近年來遺傳分析中傳遞連鎖不平衡檢定的發展特別著重於如何從多緊密連鎖的標識基因中有效地擷取單套型間相似訊息,單套基因共享概念是擷取這種相似訊息的主要方法。本研究主要考慮在親子四元體的資料結構下,以Tzeng et al.(2003)所定義之相似度為基礎,延續Yu et al(2005)方法,從改造相似度的想法著手,探討如何利用串連統計量來定義出單套型下傳組及單套型未下傳組相似度,並利用排列檢定進行檢定。模擬結果與Yu et al.(2005)比較,顯示本研究之結果所提方法之檢定力在許多情形下表現優越。
In recent years the development of transmission /disequilibrium test (TDT) in linkage analysis has been focused on how to extract haplotype similarity information from multiple closely linked marker. Haplotype sharing idea is the major method to extract such a type of information. In this thesis, following the methods of Tzeng et al.(2003) and Yu et al.(2005), we propose to use the run statistic to extract similarity information in transmitted haplotypes and non-transmitted haplotypes of case–parent tetrad families. Permutation test is used for test of association between the disease and the markers.The simulation results show that our method has higher power performaces than those of Yu et al.(2005) in some situations.
