飼料中添加樟芝固態培養基粗萃取物對點帶石斑成長表現與非特異性免疫之影響

摘要


本實驗將樟芝固態培養基以不同方式萃取以0.75%劑量添加於飼料中,探討對點帶石斑魚成長表現與非特異性免疫的影響。測定增重率(percent weight gain, PWG)與飼料效率(feed efficiency)作為成長表現指標;非特異性免疫影響測定超氧陰離子(superoxide anion,02-)誘發率、血清溶菌酶(serum lysozyme activity)與性鹼性磷酸酶(alkaline phosphatase, ALP)活性,血清生化指標測定三酸甘油脂(triglyceride)與膽固醇(cholesterol)含量。 實驗共分為六組分別於飼料中添加樟芝固態培養基甲醇、乙醇、乙酸乙酯、水粗萃取物(MAc1、MAc2、MAc3、MAc4)、直接添加組(MAc5)與不添加對照組,點帶石斑魚初始體重約9.65克,隨機撈取分組,每組12尾三重複,飼養五週,第三週與第五週記錄生長表現,第五週測定非特異性免疫與血清生化指標。 實驗結果顯示,在成長表現上飼料中添加樟芝固態培養基實驗各組增重率與飼料效率數值均高於對照組,飼料中添加0.75%樟芝固態培養基水粗萃取物有最高增重率;飼料中添加樟芝固態培養基乙酸乙酯和水粗萃取物有最高飼料效率。超氧陰離子測定結果,添加樟芝固態培養基實驗各組均高於對照組,添加水粗萃取物實驗組具有最高誘發率(0.64±0.22)顯著高於對照組(0.09±0.08)。血清溶菌酶活性測定結果顯示,實驗各組數值均高於對照組(30.78 ±3.88),添加樟芝固態培養基乙醇粗萃取物和水粗萃取物溶菌酶活性顯著高於對照組,水粗萃取物實驗組具有最高活性(52.15±2.03)。

This research studied the dietary effect of crude extract of Antrodia cinnamomea solid medium 0.75% enriched diet on the growth, non-specific immune parameters and blood biochemical indices of orange spotted grouper (Epinephelus coioides). Non-specific immune parameters include superoxide anion production ratio and serum lysozyme activity. In present study, the different extraction of Antrodia cinnamomea solid medium was supplemented 0.75% to each basal diet. Each diet was fed to triplicate of group of fish (initial body weight: 9.65 g) for 35 days. This experiment measure growth performance of orange spotted grouper at 21 and 35 day and non-specific immune parameter at 35 day. In growth performance, the results show that the feed efficiency and weight gain of orange spotted grouper fed Antrodia cinnamomea solid medium crude extract containing diets have better performance than control group, and Antrodia cinnamomea hot-water extract (MAc4) is highest. The results of non-specific immune parameters show that the production ability of superoxide anion and lysozyme activity are higher in fish fed diets supplemented with hot-water extract (MAc4) than other treatments, significantly increased compared to control(p<0.05).
