比較新型鈣矽填充材料與三氧礦化合物對骨肉瘤細胞及巨噬細胞發炎反應的影響

Comparison of inflammatory reaction of novel Ca-Si root-end filling and ProRoot MTA material on MG63 and RAW264.7 cell lines

摘要


許多研究證實,在牙科治療上作為封閉牙根尖端和其周圍組織的填充材料 Mineral Trioxide Aggregate (MTA),其生物相容性優於其他牙科填充材料。新型鈣矽填充材料與 MTA 擁有相似的物理性質,且改善了 MTA 硬化時間過長的缺點。本篇主要目的是比較 calcium silicate cement (CS) 與 MTA 對人類骨肉瘤細胞株 MG63 以及小鼠巨噬細胞株 RAW264.7 的發炎現象。選取三種濃度的 CS 與 MTA 材料萃取液,以粒線體呈色反應 (MTT) 評估萃取液對 MG63 以及 RAW264.7 的細胞毒性,再選取不會對細胞造成傷害的 10 mg/mL 材料萃取液,分別培養不同時間點的 MG63 及 RAW264.7 細胞株,利用西方墨點分析 (Western blotting) 觀察細胞中環氧酶-2 ( COX-2 ) 和誘導型一氧化氮合成酶 ( iNOS ) 蛋白質的表現量。結果顯示在 MG63 細胞株的部分,培養於 MTA 萃取液中,一天和三天後細胞的 COX-2 蛋白質表現與對照組相比有顯著性增加的趨勢 (P < 0.05),而在 CS 組別則到第五天 COX-2 蛋白質才有明顯增加的趨勢 (P < 0.05),但 CS 和 MTA 在一、三及五天彼此之間無顯著差異 (P > 0.05),對於 iNOS 分析則發現 MG63 無明顯表現。對 RAW264.7 細胞結果,兩材料皆不會誘發 COX-2 和 iNOS 表現。本實驗結果證實 CS 的發炎表現與 MTA 相似,甚至在發炎初期 CS 造成的發炎表現比 MTA 更微弱。

Mineral Trioxide Aggregate (MTA) has shown good biocompatibility and is superior to other current commercial root-end filling materials which possesses the same physic property as MTA, calcium silicate cement (CS), developed in our laboratory was under investigation in the present study. Therefore, the aim of this study was to compare CS to MTA in their abilities of inducing cellular inflammatory reactions of MG63 and RAW264.7 cell lines. MG63 cells were incubated in the eluate of 10 mg cement per ml medium for 1, 3, 5, and 7 days, respectively, and then cells were harvested and examined for their Cyclooxygenase-2(COX-2) and nitric oxide synthase (iNOS) protein expressions using Western blot technique. A similar procedure was performed on RAW264.7 cells except that the harvest time points were 1, 12, and 24 hr. Both cements induced COX-2 expression in MG63 cell line, but not iNOS expression. COX-2 expression was higher on days 1 and 3 by MTA and CS triggered higher expression on day 5 (P < 0.05). On the other hand, both materials did not induce COX-2 and iNOS expressions in RAW264.7 cells. This experiment show that CS and MTA have similar inflammatory effect , even the effect of CS is unobvious compare by MTA in the early stage.
