液相層析串聯質譜技術應用於毛髮中常見濫用藥物之分析與探討

摘要


目前國內藥物濫用的情形日益嚴重,青少年更成為毒品販售的主流。毛髮為新興的生物檢體,可記錄受檢者服用藥物的歷程,根據採樣位置,可追溯過去數月甚至數年之藥物史,因此毛髮檢驗在鑑識科學上得到極大的重視。然而,毛髮檢驗目前主要之分析方法:氣相層析質譜儀(GC/MS)之樣品前處理步驟複雜,且需要針對其分子特性與特殊官能基來進行不同的衍生化;相較之下,液相層析質譜儀(LC/MS),則有簡單、快速、不需經衍生化步驟即可達到精確分析的目的。 本研究主要是發展LC/MS之毛髮檢驗方法,針對常見濫用藥物:甲基安非他命、安非他命、MDMA、MDA、MDEA、6-乙醯嗎啡、可待因、嗎啡、K他命和古柯鹼,進行之方法開發。實驗首先以0.1 M Phosphate buffer(pH 5.0)浸泡毛髮,隨後進行SPE萃取,並以C18的管柱進行分離,接著以LC/MS/MS (API3000)進行MRM模式之偵測。實驗結果發現:除可獲得良好的分離效果外,LC/MS/MS的靈敏度較GC/MS有更低的方法偵測極限。最後,我們以LC/MS/MS分析真實檢體,並利用LC/MS/MS不需衍生化之優點,排除GC/MS分析服用假麻黃素藥物可能造成的誤判。

Drugs of abuse have become a critical issue in the country and the adolescents are reported to be the main population involved. Hair is a new alternative biological specimen, get great attention in the forensic science in recent years because its ability of trace back detection. However, current available analysis method in hair testing, gas chromatography-mass spectroscopy (GC/MS), includes considerably complex processes for the sample preparation. GC/MS requires various derivations for the analysis based on certain compound characteristics and specific functional groups. Compared to GC/MS, liquid chromatography-mass spectroscopy (LC/MS) provides a more simplified, effective way to achieve accurate analysis purpose without any sample preparation for the derivation. In this study, the hair testing using LC/MS has been developed to detect methamphetamine, amphetamine, MDMA, MDA, MDEA, 6-monoacetylmorphine, codeine, morphine, ketamine, and cocaine. For the experimental procedures, the hair was soaked in 0.1 M phosphate buffer (pH 5.0), and processed through SPE extraction method. The sample was then injected into C18 column for the species separation and analyzed by LC/MS/MS (API3000) under multiple reaction monitoring (MRM) mode. The results indicate this method can provide high separation efficiency for those species and the LC/MS/MS can provide the greater limit of detect(LOD) than GC/MS. Finally, a real hair sample was applied to verify this methodology. The advantage of LC/MS/MS is that the derivation is not required which can eliminate possible false negative results by GC/MS from consumer of pseudoephedrine .
