染布青萃出物抑制雞卵白蛋白致敏氣喘小鼠呼吸道過度反應與發炎

摘要


過敏性氣喘是一個呼吸系統疾病,其特徵有呼吸道過度反應(hyperresponsiveness,AHR)、支氣管阻塞、氧化壓力與發炎反應。在大部分的氣喘患者,肺部過度活化Th2細胞免疫反應,且當過敏原增加時,異常活化Th2細胞,增加細胞激素如IL-4與IL-13的分泌,影響氣喘的生理病理發展。IL-4與IL-13可活化B細胞並促進免疫球蛋白IgE的產生,而增加肺部發炎與呼吸道過度反應。染布青是傳統中草藥,可用於治療發炎症狀,因此在實驗中探討染布青萃出物調控雞卵白蛋白(ovalbumin,OVA)致敏氣喘小鼠模式中所誘導的Th2細胞免疫反應與發炎之功效。研究結果顯示,OVA致敏氣喘小鼠,給予染布青水萃與乙醇萃顯著減緩AHR,並且降低IgE的產生,減少發炎細胞浸潤、氧化壓力與肺沖洗液中IL-13及TNF-α的表現。此外,染布青萃出物降低肺部組織細胞內訊息路徑MAPKs的磷酸化,進而降低轉錄因子(nuclear factor-kappa B,NF-κB)進入細胞核內與NF-κB結合DNA的活性,可抑制細胞激素IL-13與TNF-α產生,減緩肺部發炎反應。以上結果顯示,染布青萃出物抑制OVA誘發過敏性氣喘發生,且染布青萃出物可經由MAPKs與NF-κB路徑減緩過敏性氣喘所造成的肺部發炎反應。

Asthma is a chronic respiratory disease characterized by airway hyperresponsiveness (AHR) and obstruction as well as pulmonary inflammation and oxidative stress. In the majority of asthmatics, excessive activation of T helper 2 (Th2) cells occurs in lungs. Aberrant activation of Th2 cells by allergens increased secretion of cytokines such as interleukin (IL) -4 and IL-13 which are crucial for developing the pathophysiological symptoms of asthma. IL-4 and IL-13 plays a role in activation of B cells for immunoglobulin (Ig) E synthesis, increase of inflammatory responses and AHR. Indigofera suffruticosa Miller is a herbal medicine for the treatment of inflammation. In this study, we investigated the effect of I. suffruticosa extracts on asthmatic responses in the Th2-mediated ovalbumin-induced allergic asthma model. Data show that tretment with I. suffruticosa water and ethanol extracts significantly attenuated AHR as measured by the enhanced pause response. Moreover, I. suffruticosa extracts diminished oxidative stress, IgE secretion, inflammation cell infilitation, levels of IL-13 and TNF-α in bronchoalveolar lavage fluid. Indigofera suffruticosa extracts reduced mitogen-activated protein kinases phosphorylation and nuclear factor-kappa B (NF-κB) nuclear translocation and NF-κB nuclear protein DNA binding activity in lung tissues.
