菊糖於STZ誘發之小白鼠糖尿病之影響

Effects of Inulin in STZ-induced Diabetes mice

摘要


糖尿病已為台灣地區最重要的慢性病之一,罹患率高居國人十大死因之一。糖尿病所引起的併發症為病患死亡的主要原因,糖尿病併發症的發生與糖尿病所導致的血糖過高(hyperglycemia)有密切相關。在體內過高的血糖濃度,細胞內的葡萄糖會經由多元醇(polyol pathway)來進行代謝,許多文獻指出此代謝路徑會導致糖尿病併發症-視網膜病變、神經病變以及心血管疾病之發生,再者糖尿病者常伴隨著脂質代謝異常,高血糖的氧化壓力是誘發血管病變的關鍵因子,因此良好的血糖控制對於糖尿病患減緩併發症的發生是很重要的。 本研究探討菊糖於streptozotocin (STZ)誘發小鼠糖尿病模式之影響。使用BALB/c 雄鼠經由尾靜脈注射STZ (200mg/kg BW)誘發糖尿病,菊醣是一種含有大量的果寡糖的天然物,而果寡糖具有膳食纖維特性,因此本實驗目的在探討菊糖添加入飼料中,對STZ誘發之糖尿病小鼠體內脂質及醣類代謝之影響。實驗共進行四週,分為四組(1)Control組 (2)Diabetes組 (3) Diabetes+inulin組 (4) Control+inulin 組,實驗期間給予每隻小鼠每日5克飲食攝取量並添加濃度11%的菊醣及自由飲水。 結果顯示,食用添加菊糖的糖尿病鼠,降低了血漿及肝臟中醛糖還原酶、乙二醛酶的活性與肝臟中山梨醇脫氫酶的活性,而血漿中的葡萄糖、三酸甘油脂、膽固醇及纖維網狀蛋白的量都有明顯下降,在抗氧化作用的部分,食用添加菊糖的糖尿病鼠,血漿及肝臟中丙二醛值有顯著下降,而麩胱酐肽氧化酶濃度則顯著上升。上述實驗結果顯示菊糖應具有預防或降低糖尿病併發症發生的效果。

Diabetes is one of the most important chronic diseases in Taiwan. The complications from diabetes mellitus are the main causes responsible for the death of patients. Hyperglycemia occurred in diabetes results in oxidative and glycative stress from polyol pathway. Many papers have indicated that both oxidation and glycation contribute to the development of diabetic neuropathy, diabetic nephropathy and cardiovascular disease. Thus, any agents with anti-oxidant and/or anti-glycative effect may retard the presence of diabetic complications. Inulin is a natural compound with fiber-like property. In our present study, streptozotocin (STZ) was used to induce diabetes in Balb/c mice, and inulin was supplied. The purpose of this study was to investigate the effect of this compound on anti-oxidant protection and lipid metabolism in diabetic animal model. Results showed that the intake of inulin decreased the activity of aldose reductase, glyoxalase I and sorbitol dehydrogenase in the plasma and liver. Inulin intake also increased glutathione peroxidase activity, reduced lipid oxidation level and decreased the level of blood glucose, triglyceride, cholesterol and fibronectin in diabetic mice. These results suggested that inulin might provide alleviative effects against diabetic deterioration.
