麻芛抗發炎活性之研究

Study on the antiinflammatory effects of Corchorus olitorius L.

摘要


麻芛生長於臺灣夏季,日本、埃及等,世界各地也都有栽種。素有皇室蔬菜「moroheiya」的美稱。在臺灣,食用前先以清水搓揉沖洗以去除苦味,並與地瓜和小魚乾一同煮成羹湯的方式食用。 在預備試驗中,將麻芛分為全葉(whole leaves)、去莖葉(leaves)、莖(stem)、水洗葉(washed leaves, WL)與水洗液(washed water,WW)等五種區分物之酚類萃取物。除了WL與WW之外,不論是清除DPPH自由基的能力、抑制脂質氧化能力(linoleic acid emulsion system)或是螯合亞鐵離子的能力,其他三種酚類萃取物皆具有統計上顯著高於WL與WW的抗氧化能力。在發炎調節的功效上,除了WL與WW之外,也可以發現不論是抑制NO與PGE2的生成,以及抑制iNOS與COX-2 的蛋白質表現上,其他三種萃取物皆由具有統計上顯著地抑制發炎反應的功效。而且不論是在抗氧化或發炎調節的功效上,尤其是全葉的酚類萃取物效果更佳。 本研究以麻芛全葉分別以不同極性之溶劑將麻芛全葉分成五種不同極性萃取物,分別為n-hexane、ethyl acetate (EtOAc)、acetone、methanol、water extracts。結果顯示,不論是清除DPPH自由基的能力或是抑制脂質氧化的能力(linoleic acid emulsion system),EtOAc、acetone及methanol extracts皆具有統計上顯著的抗氧化能力。而在發炎調節的功效上,不論是抑制PGE2的生成或是抑制COX-2的蛋白質表現上,EtOAc、acetone及methanol extracts也皆具有統計上顯著的抑制發炎反應的能力。而且在三種萃取物中皆發現有高量的chlorogenic acid、caffeic acid 及 myricetin,更在EtOAc與acetone extracts中發現一種未知的化合物。因此,chlorogenic acid、caffeic acid、myricetin與未知的化合物或許在抗氧化與發炎調節上扮演重要的關鍵角色。在未來可以朝找出麻芛中關鍵生物活性成份著手,進一步找出與癌症化學預防的相關性。

Corchorus olitorius L. is grown in Taiwan during summer. Tender leaves are crushed and washed by running water before cooking. In preliminary study, five fractions including crude phenolic extracts (using 80 per cent aqueous acetone) of whole plant, leaf, stem, washed leaf (WL) and dried water washing material (WW) were used in this study. Except for WL and WW, the phenolics of jute of all fraction showed the good antioxidative activity and the regulation of inflammatory responses, and the whole plant had the best effect. In the second study, five solvent extracts of tender leaves of Corchorus olitorius L. including n-hexane, ethyl acetate (EtOAc), acetone, methanol and water extracts, were used in this study. Acetone extracts showed better antioxidative activity and the regulation of inflammatory responses. Ethtlyl acetate (EtOAc) and methanol extracts also showed good inhibitory effects on inflammatory responses High levels of chlorogenic acid, caffeic acid and myricetin were detected in EtOAc and methanol extracts of Corchorus olitorius L. caffeic acid and an unknown compound were also detected in acetone and EtOAc extracts. This meaned that chlorogenic acid, caffeic acid, myricetin and the unknown compound A may play an important role on inflammatory regulation. The further study of the main contributors to antioxidation and inflammatory inhibition are required in future.
