表兒茶素沒食子酸酯抑制卵巢癌腫瘤生長能力之研究

Study of the Inhibition Effect of Epicatechin Gallate on Cell Growth in Ovary Cancer

摘要


卵巢癌 (Ovary cancer;OC) 發生率在全球位居第七位。卵巢癌發生在初期被發現是不容易的,而且患者本身也很少會去注意,因此當被發現時大多已經是第 3 期或是已經到達第 4 期了,缺乏有效的篩檢工具,縱使經由手術切除或放射治療後,近 90% 的人會復發依舊難以根治。癌症的形成錯綜複雜,真正的成因至今還是個謎,高額的標靶藥物與漫長的療程對患者本身也是另一種煎熬,所以尋求更簡單易取得,且對身體其他正常細胞沒有毒性的天然抗癌藥物,是目前最熱門的議題之一。表兒茶素沒食子酸酯(Epicatechin gallate ; ECG),在綠茶內的生物活性佔有高成分,已有報告指出可抗氧化、清除自由基、抗過敏、改善心血管疾病、以及抗發炎也可以調節免疫系統的作用。本實驗研究表兒茶素沒食子酸酯是否能抑制人類卵巢癌細胞的生長,以 MTT assay與 colony formation assay,研究結果證實表兒茶素沒食子酸酯能抑制細胞存活率 (cell viability) 及降低 colony 的形成,再以免疫缺陷小鼠 (BALB/cAnN.CgFoxnlnu/CrlNarl) 進行動物實驗,觀察皮下注射卵巢癌細胞 (PA-1) 後,結果顯示老鼠體重沒有明顯差異,利用活體影像分析系統 (In vivo imaging system;IVIS) 觀察腫瘤在活體內的生長狀況,結果顯示體表腫瘤生長和腫瘤組織大小,給予表兒茶素沒食子酸酯 20 mg/kg 劑量的組別腫瘤生長有明顯被抑制;最後把老鼠犧牲後將腫瘤取下,與控制組相比在表兒茶素沒食子酸酯20 mg/kg 劑量的組別腫瘤的重量也有明顯被抑制。總結以上表兒茶素沒食子酸酯對卵巢癌腫瘤 (PA-1) 腫瘤生長是有抑制的效果。

Ovary cancer has the seventh highest incidence in the world, Ovary cancer is not easy to detect in its early stages and is rarely noticed by the patients themselves, so by the time it is detected it is usually already at stage 3 or stage 4. In the absence of effective screening tools, even after surgical removal or radiotherapy, nearly 90 % of recurrences remain incurable. The true cause of cancer is still a mystery, and the high cost of targeted drugs and lengthy treatment courses are another source of suffering for patients themselves. The search for natural anti-cancer drugs that are easier to obtain and not toxic to other normal cells in the body is one of the hottest issues at the moment. Epicatechin gallate (ECG) is the high bioactive content of green tea has been reported to have antioxidant, free radical scavenging, anti-allergy, cardiovascular disease improving, anti-inflammatory, and immune system modulating effects. In this experiment, we investigated whether ECG could inhibit the growth of human ovary cancer cells by MTT assay and colony formation assay. The results of the animal experiment with immunodeficient mice (BALB/cAnN.CgFoxnlnu /CrlNarl), observed after subcutaneous injection of ovary cancer cells (PA-1), showed no significant difference in body weight. In vivo imaging system (IVIS) is used to observe the growth of tumor in vivo, which shows the growth of tumor and the size of tumor tissue on the body surface. Tumor growth was significantly inhibited in the group given ECG at a dose of 20 mg/kg. Finally, tumor weight was also significantly inhibited in the group given ECG at a dose of 20 mg/kg compared to the control group when the mice were sacrificed and the tumor were removed. In conclusion, ECG has been shown to have an inhibitory effect on the growth of ovary cancer tumor (PA-1).
