乳癌中 COX-2 與 c-Src 蛋白表現量增加且具依存性

Concomitant enhancement of COX-2 and c-Src in breast carcinoma

摘要


iNOS、COX-2 和 c-Src 過度表達於惡性腫瘤中,其中包含乳癌,但其調控機轉尚不清楚。本篇研究主要在比較 iNOS、COX-2 和 c-Src 的表現在乳癌細胞與周圍的正常基質細胞之間的差異性,以及比較 iNOS、COX-2 和 c-Src 的表現與年齡、腫瘤大小、局部淋巴結浸潤、組織型態分級、細胞核分級、臨床分期、C-Erb B2、ER、PR 表現及 iNOS、COX-2 與 c-Src 三個蛋白彼此之間表現的相關性。我們收集了 24 例乳癌病理組織切片進行免疫組織化學染色來測定此三種蛋白的表現。iNOS、COX-2 和 c-Src 在乳癌細胞的表現量明顯高於在基質細胞的表現量 (p < 0.001)。COX-2 和 c-Src 的表現與乳癌臨床分期呈負相關性 (p = 0.034, p = 0.029) 且 COX-2 蛋白與 c-Src 蛋白表現呈正相關性 (p = 0.020)。由這些結果看來乳癌中 COX-2 與 c-Src 皆過量表現且彼此含量具依存性;此外, COX-2 與 c-Src 的過度表達可能與較不具侵犯性的乳癌表現型有關。

Inducible nitric oxide synthase (iNOS), Cyclooxygenase-2 (COX-2) and c-Src are overexpressed in malignant tumours including breast cancer, though the mechanisms of their upregulation are unclear. This study aimed to determine the expression of iNOS, COX-2 and c-Src in breast cancer cells in comparison to the surrounding normal stromal cells. At the meantime, the relationship of iNOS, COX-2 and c-Src expression in breast tumor specimens with age, tumor size, regional lymph nodes infiltration, histological grade, nuclear grade, TNM stage, C-Erb B2 expression, estrogen receptor (ER), progesterone receptor (PR) was examined. The amount of iNOS, COX-2 and c-Src was determined by immunohistochemical staining in 24 tissue sections collected from breast cancer patients. iNOS, COX-2 and c-Src immunoreactive score was significantly higher in the breast cancer cells than in the stromal cells (p < 0.001). COX-2 and c-Src expression was inversely correlated with TNM stage (p = 0.034, p = 0.029) and COX-2 expression was positively correlated with c-Src expression (p = 0.020) in breast cancer cells. Taken together, we observed the concomitant enhancement of COX-2 and c-Src in breast carcinoma and their simultaneous overexpression might be related to the progression of breast carcinomas with low aggressiveness.
