蘑菇對於細菌內毒素誘發氧化壓力動物模式抗氧化能力之效用

The effects of the Antioxidant capacity of Agaricus bisporus (mushroom) on mice after given lipopolysaccharide

摘要


近年來,由於國人糖尿病、肥胖、癌症、心臟血管疾病的比例上升,而對於這些慢性病被認為最有效的預防方法之ㄧ是均衡飲食。本研究是在探討台灣蘑菇(Agaricus bisporus;AB)以10%和20%不同濃度與AIN-76飼料一起混合飼育小白鼠,觀察其對於血糖調控、抗發炎及抗氧化能力的影響。 ICR雄鼠經由上述方式飼育經過一個月,經腹腔注射細菌內毒素( Lipopolysaccharide;LPS,280μg )誘發氧化壓力,其組別分別為(1)Control(2)LPS(3)10%AB(4)LPS+10%AB(5)20%AB(6)LPS+20%AB。結果顯示,食用蘑菇的組別,其血糖濃度及葡萄糖六磷酸酶(Glucose-6-phosphatase;G6Pase)活性均顯著高於LPS組,有助於血糖的回升。而血清中乳酸去氫酶(Lactate dehydrogenase;LDH)、纖維網狀蛋白(Fibronectin;FN)和TNF-α均顯著低於LPS組。蘑菇的補充顯著增加感染LPS的小鼠其肝臟中過氧化氫酶(Catalase;CAT)的活性並降低脂質過氧化作用;而且顯著增加感染LPS的小鼠其血清中麩胱苷肽過氧化酶(Glutathione peroxidase;GPx)的活性及麩胱苷肽(Glutathione;GSH)。蘑菇的補充使感染LPS的小鼠其血清及肝臟中超氧歧化酶(Superoxide dismutase;SOD)的活性顯著上升。上述結果顯示,台灣蘑菇在氧化壓力下可調控血糖、預防發炎及氧化的發生。

This study was to investigate the effect of Taiwanese Agaricus bisporus intake of different concentrations on glycaemic control, anti-inflammation and antioxidant capacity in mice. Agaricus bisporus powder at 10% and 20% were added into the diet for 1 month. Mice were treated with lipopolysaccharide to induce oxidative stress. Results showed that the intake of Agaricus bisporus significantly rose glucose level and liver glucose-6-phosphatase activity but significantly reduced lactate dehydrogenase activity , fibronectin and tumor necrosis factor-alpha levels in serum (p<0.05). These supplements significantly increased liver catalase activity and decreased lipid oxidation (p<0.05). The administration of both 10% and 20% Agaricus bisporus significantly increased glutathione peroxidase (GPx) activity and glutathione (GSH) level (p<0.05) in serum. However, only 20% Agaricus bisporus treatment significantly increased GPx activity and GSH level (p<0.05) in liver. The administration of both 10% and 20% Agaricus bisporus significantly increased superoxide dismutase (SOD) activity (p<0.05) in serum and liver. These data suggest that the Agaricus bisporus intake could maintain glucose homeostasis, decrease inflammation status, and enhance antioxidant protection.
