洛神花各成分萃取物對於黑色素瘤細胞黑色素生成之影響

摘要


隨著社會的進步,經濟的富裕,美容醫學的市場可說是發展迅速,潛力無限,人們對於美白的追求更沒有年齡、階層的界線,幾乎已成全民運動。許多專家學者對各種天然物的萃取物在抑制黑色素形成及去斑美白的研究,更是積極的研究發展,這同時也是目前化妝品開發及治療上的主流。 本研究主要探討洛神花各成分之萃取物( Hibiscus sabdariffa extracts;HSE )對於黑色素瘤細胞( B16-F1 )中黑色素生成的影響,利用抑制酪胺酸酵素( Tyrosinase )活性試驗及細胞培養試驗,將不同的洛神花萃取物進行抑制細胞內黑色素含量及酪胺酸酵素活性抑制能力的評估,找尋安全有效的美白製劑。結果顯示洛神花各成分之萃取物對於培養中之細胞生長密度均無影響,且顯示我們選用之劑量並不會對於黑色素瘤細胞產生細胞毒性。在細胞內黑色素含量測定試驗中,發現洛神花各成分之萃取物均有顯著地降低了細胞內的黑色素含量,另外在測定酪胺酸酵素活性實驗中,不論是利用分光光度計或是活性膠體試驗,皆可觀察到洛神花各成分之萃取物對於細胞內的酪胺酸酵素活性具有不同的抑制能力。由本實驗結論可推知,洛神花萃取物能夠進一步發展為有效且安全的美白成分,但其詳細的機轉則值得進ㄧ步的研究。

Our society has been constantly developed and economic status has been quickly enhanced, which helps create a fast blooming cosmetic surgery market. There is no limitation for people; regardless of their social status or age difference, pursuing whiter and fairer skin. Many experts conduct researches about spots removing and natural ingredient extracts application to eliminate Melanie forming. Such research becomes the current dominant treatment of cosmetic and therapeutic treatment development. This search aims to discuss the cause and effect relationship between Hisbiscus sabdarfiffa extracts( HSE ) and melanin that is generated from human melanoma cells. By conducting the clinical experiment of inhibits tyrosinase activity and culturing cell samples, to evaluate the ability of controlling melanin quantity contained within cells and inhibits Tyrosinase activity once different types of Hibiscus sabdariffa extracts are added, to discover safe and effective topical skin-whitening agents. The result shows there is no determined influence upon the growing rate and density of the cells. It also shows the dosage we apply won’t cause toxic effect in melanoma cells. During the test of melanin quantity contained within cells, it is found that HSE extracts have distinctively lower down the melanin quantity contained within cells. In addition, during the experiment of tyrosinase clinical experiment; either by applying spectrophotometer or Tyrosinase zymorgraphy test ; which leads to a finding that HSE have different levels of elimination power to Tyrosinase within cells. This study indicates Hisbiscus sabdarriffa extracts can be further developed as safe and effective whitening products. However, for any further finding, more advanced researches have to be conducted to justify it.
