在過去的實驗中證明七層塔水萃取物有著抑制肝臟星狀細胞活化的功效,而在四氯化碳誘導大鼠肝纖維化的動物試驗模式中,也發現七層塔純化物有著抑制肝臟纖維化的功效。本實驗目的為測試七層塔(Ocimum gratissimum)萃取粉 10% 對肝功能功效性的評估。首先進行動物亞急毒性試驗,以不同劑量的七層塔萃取粉,實驗操作4組,連續管餵28天後觀察實驗組與對照組的差異,結果顯示七層塔萃取粉10% 是安全無虞的。再來測試七層塔(Ocimum gratissimum)萃取粉10% 之肝臟的保護的效果,實驗操作5組,實驗組腹腔注射8% CCl4 (0.1 ml/100 g BW),並分別灌餵予Silymarin、低劑量和高劑量的七層塔萃取粉10%,共為期八週,結果顯示經過七層塔萃取粉處理能預防四氯化碳所誘導的肝臟細胞型變產生,減少細胞間質膠原蛋白堆積;血清Alanine transaminase (ALT)和Aspartate transaminase (AST)活性下降;同時回復抗氧化酵素活性和抗氧化成分。透過大鼠肝纖維化實驗,確認了七層塔萃取粉10% 能有效保護肝臟,提高抗氧化酵素並降低自由基的傷害,而抑制肝臟的纖維化。雖然七層塔萃取粉10% 並沒有完全恢復四氯化碳所誘導的肝臟細胞變型產生,但是200 mg/Kg七層塔萃取粉抑制的效果幾乎與200 mg/kg silymarin所造成的效果相同。綜合以上結果,推估七層塔萃取粉10% 能預防肝臟發炎及抑制肝臟纖維化。
In the previous study, we demonstrated that Ocimum gratissimum could inhibit HSC activation. Moreover, the purified Ocimum gratissimum also inhibited the CCl4-induced liver fibrosis.The purpose of this request research program is to determine the hepaprotective effect of Chhit-Chan-Than extract powder 10% (Ocimum gratissimum). First, we determined the toxicity of Chhit-Chan-Than extract powder 10%. After 28 days of continuous tube feeding with different dose of Chhit-Chan-Than extract powder 10%, the results of biochemistry、urine biochemistry and hematology were no change. Therefore, we suggested that Chhit-Chan-Than extract powder 10% was safety in vivo. Furthermore, we investigated the hepaprotective effects of Chhit-Chan-Than extract powder 10% in vivo. The results indicated that the CCl4-induced histopathogical changes may be prevented by the Chhit-Chan-Than extract powder 10% through reducing the intercellular collogen stack, dropping blood serum ALT and AST levels; and replying the catalase activity and GSH content (the anti-oxidative enzyme activity and the anti-oxidative ingredient). Although Chhit-Chan-Than extract powder 10% has not completed to resume the formation of liver fibrosis induced by CCl4, the suppression of Chhit-Chan-Than extract powder 200 mg/Kg was nearly similar to the silymarin. Take together; we suggested that the Chhit-Chan-Than extract powder 10% was able to prevent the liver inflammation and fibrosis.
