白藜蘆醇抑制TPA誘發基質金屬蛋白酶-9與口腔癌轉移機制之探討

在台灣,口腔癌的發生率以及死亡率日漸升高,而在口腔癌患者中,有超過90%是屬於鱗狀細胞癌 (Squamous cell carcinoma, SCC)。儘管在不斷進步的口腔癌治療其中包括手術、放射與化學療法治療下,口腔癌患者的五年存活率也維持在約50%。研究指出,化學預防也許是一個治療口腔癌的方向。白藜蘆醇 (resveratrol) 是一種非黃酮類的酚類物質,其療效具有抗氧化、保護心血管、保護神經等功能,在過去文獻指出,白藜蘆醇具有抗乳癌細胞轉移的效果,但白藜蘆醇在口腔癌的相關轉移機制則尚未明確。本實驗目的擬探討白藜蘆醇對於抑制口腔癌細胞株SCC-9機制,我們利用MTT assay偵測細胞存活率,接著使用migration assay、Wound-Healing assay、Zymography、reverse-transcriptase polymerase chain reaction (PCR)以及promoter assays證實白藜蘆醇藉由抑制MMP-9的分泌與蛋白表現來降低癌細胞的轉移與侵襲。我們證明了白藜蘆醇在0~100

Oral cancer exhibits the highest rate of increasing incidence and mortality of all cancers in Taiwan. Squamous cell carcinoma (SCC) represents more than 90% of all oral malignancies. Regardless of improvements in therapeutic applications, the 5-year survival rate for patients with advanced diseases after multimodality treatment, including surgery, radiotherapy, and chemotherapy, remains approximately 50%. Research has indicated that combination of chemoprevention might be a more practical approach to treating oral cancers. Resveratrol is a stilbenoid, a type of natural phenol, that has antioxidative, cardioprotective, and neuroprotective properties. The literature has reported that resveratrol can inhibit cell metastasis in breast cells. However, the effect of resveratrol exerts on the metastasis of oral cancer cells remains unclear. In this study, we investigated the effect the anti-invasive activity of resveratrol on a human oral cancer cell line (SCC-9) in vitro and the underlying mechanisms. Cell viability was examined by MTT assay, whereas cell motility was measured by migration and wound-healing assays. Zymography, reverse-transcriptase polymerase chain reaction (PCR), and promoter assays confirmed the inhibitory effects of resveratrol on matrix metalloproteinase-9 (MMP-9) expression in oral cancer cells.
