迷迭香酸抑制塵螨過敏原Der p 2誘發人類肺纖維母細胞MRC-5前纖維化基因表現之研究

摘要


由塵螨所引起的過敏是國人普遍常見的免疫性疾病,台灣常見的塵螨過敏來源有三種:歐洲室塵螨、美洲室塵螨與熱帶無爪室塵螨,其中歐洲室塵螨是台灣數量最多、最重要的塵螨過敏來源。先前的研究顯示,塵螨所引起的過敏若嚴重程度較高可能會導致肺纖維化的進行。本研究利用不具有蛋白水解酶活性的歐洲室塵螨第二群過敏原Der p 2刺激人類肺纖維母細胞株MRC-5 24小時後,觀察三個纖維化指標基因的表現量,發現α-平滑肌肌動蛋白(α-SMA)、纖維連接蛋白(fibronectin)及第一型膠原蛋白(collagen I) mRNA的表現量有明顯增加。迭香酸是一種具強抗氧化性的酚酸類化合物,具有抗發炎、抗過敏、抗肝纖維化等生物活性。我們發現使用迷迭香酸預先處理過人類肺纖維母細胞後,再以Der p 2刺激細胞,然後利用反轉錄-聚合酶鏈反應(RT-PCR)跟即時定量聚合酶鏈鎖反應(Real-time quantitative RT-PCR)等分析方法,觀察其三個前纖維化基因的表現,發現迷迭香酸可以降低Der p 2所誘導的α-平滑肌肌動蛋白、纖維連接蛋白及第一型膠原蛋白的mRNA表現。接著再利用免疫轉漬(western blot)分析方法,觀察相關的訊息路徑包括AKT、ERK1/2、JNK、P38等蛋白的磷酸化,試著釐清迷迭香酸抑制Der p 2所誘導纖維化的路徑。結果發現迷迭香酸具有抑制Der p 2所誘導的AKT磷酸化,但是對ERK1/2、JNK、P38等蛋白激酶的磷酸化並無顯著影響。綜合以上發現,我們認為迷迭香酸可能是透過抑制AKT的訊息路徑,進而抑制MRC-5細胞中前纖維化基因的mRNA表現,推測迷迭香酸可能具有抑制肺臟纖維化及預防因塵螨引起的呼吸道過敏導致纖維化的功效,未來可進一步評估其臨床治療與預防保健的效果。

House dust mites (HDMs) are the major cause leading to airway hypersensitivity in Taiwan and developed countries.In Taiwan, ther are three main species of HDMs leading to allergic disoders,includingDermatophagoides pteronyssinus, Dermatophagoides farina, and Blomia tropicalis. Among the HDMs, pathogenic mechanisms of Dermatophagoides pteronyssinus-induced allergies arewidely investigated and allergens derived from the mite are mostly identified and characterized. Previous studies have indicated that severe respiratory hypersensitiveness maycontribute to further pulmonary fibrosis.Rosmarinic acid (RA) is a phenolic compound with highly antioxidant activity andhas anti-inflammatory, anti-allergic and otherbiological activities. However, effects of RA on fibrosis remain unclear. Thus, the present study wasaimed to invesitigate effects of Der p 2, a non-proteolytic major allergen derived from Dermatophagoides pteronyssinus,on profibrotic gene expression in human lung fibroblast MRC-5.
