探討薯蕷皂苷元diosgenin對卵巢去除雌鼠脂質代謝之影響

Effects of diosgenin supplementation on lipid metabolism in ovariectomized rats

摘要


本研究主要目的是探討於飲食中補充diosgenin對於停經動物模式-卵巢去除雌鼠血液脂質、肝臟膽固醇及三酸甘油酯、糞便中性固醇及膽酸排出之影響。 96隻8週齡Wistar雌性大鼠適應四週後,於12週齡時進行偽手術(Sham組,n = 48)或去卵巢手術(OVX組,n = 48)。經過四週術後恢復期,再區分為四小組,分別餵食含不同劑量之diosgenin (0, 10, 50及100 mg/kg BW/day)麵糰四週,於20週齡時犧牲動物。實驗期間內採自由飲水及攝食;並於犧牲前三天記錄攝食量及收集糞便,犧牲後採集心臟血液及肝臟進行後續分析。 血脂方面,卵巢去除雌鼠血漿中總膽固醇與低密度脂蛋白膽固醇較偽手術組高。以單因子變異數分析後,偽手術組與去卵巢組組內補充不同劑量diosgenin,對Wistar雌鼠之血漿中總膽固醇、高密度脂蛋白膽固醇、低密度脂蛋白膽固醇、低密度脂蛋白膽固醇/高密度脂蛋白膽固醇比值及三酸甘油酯皆無任何影響。另外,卵巢去除組中,補充10、50及100 mg diosgenin/kg BW組肝臟膽固醇皆顯著低於未補充diosgenin組;然而補充不同劑量diosgenin均對肝臟三酸甘油酯無任何影響。在糞便中性固醇及膽酸排出量方面,在卵巢去除組中,補充10及50 mg diosgenin/kg BW組糞便中性固醇排出量顯著高於未補充組;並且補充100 mg diosgenin/kg BW組膽酸排出量亦顯著高於未補充組。 因此,本篇研究建議適量攝取diosgenin具有改善停經婦女體內膽固醇代謝的保健潛力。

The aim of this study was to investigate effects of diosgenin supplementation on blood lipid, liver cholesterol and triacylglycerol contents, fecal neutral sterol and bile acid excretion in ovariectomized female Wistar rats. Rats were sham-operated or ovariectomized (OVX) at age of 12 weeks. After four weeks of recovery, rats further divided into four subgroups and supplemented with 0, 10, 50 or 100 mg/kg BW/day of diosgenin for 4 weeks. The 72-h fecal samples were collected and rats were fasted overnight before sacrifice. Plasma and liver were stored in -20 C for further analyses. The plasma total cholesterol and HDL-cholesterol concentrations in OVX groups were higher than those in the sham groups, respectively. Diosgenin supplementation did not affect the plasma total, HDL-, LDL-cholesterol and triacylglycerol concentrations, and LDL/HDL-cholesterol ratio in the OVX group. However, all doses of diosgenin supplementation significantly lowered the hepatic cholesterol concentration. Moreover, the hepatic triacylglycerol concentration was not affected by diosgenin supplementation. In addition, the fecal neutral sterol excretion in the OVX rats fed with 10 and 50 mg/kg BW/day of diosgenin were significantly higher than the control group. The fecal bile acid excretions were significantly increased with 100 mg/kg BW/day of diosgenin supplementation in the OVX rats. In conclusion, the results of this study suggest that optimal diosgenin intake could improve the cholesterol metabolism in menopause women.
