蘋果多酚對於卵巢切除母鼠之燙傷修護探討

摘要


婦女在更年期時,因賀爾蒙的減少而造成皮膚老化,雌激素在此進程中扮演著重要的角色。皮膚的老化也降低皮膚傷口癒合的效率。根據先前先進學者的研究,皆有許多文獻探討老化或慢性疾病與皮膚傷口癒合相互關係的機制,但並無完全有效的治療方式。而先前研究文獻也指出蘋果多酚具有良好的抗氧化作用,並且對人體有許多的益處。因此,本實驗想要探討蘋果多酚是否會促進卵巢切除母鼠的皮膚傷口癒合。首先,以母鼠作為實驗模型,進行卵巢切除手術,模擬停經症候群之婦女,並讓母鼠於術後靜養四週,待切割之傷口復原並恢復活力後,將老鼠以氣體麻醉方式麻醉並進行背部除毛,以一平方公分之鐵塊燒熱至100℃燙出傷口,並以此燙傷模式作為本篇之研究探討。燙傷後,蘋果多酚以混入空白基質霜的方式給予皮膚傷口敷藥治療,每天換敷蘋果多酚五或十天。透過免疫組織染色結果顯示,傷口處的皮膚,脂肪細胞增加,皮膚表皮層出現缺口,毛囊排列也變得不規率,經給予蘋果多酚治療五或十天後,皮膚傷口處脂肪細胞減少,皮膚表皮層新生並修復,毛囊排列恢復規率性,這結果指出皮膚上皮再生作用的現象。細胞實驗方面,本研究使用皮膚真皮層纖維母細胞探討皮膚傷口癒合機制,細胞在給予蘋果多酚治療後,會增加爬行蛋白p-FAK、MMP-2的表現,進而證實蘋果多酚能夠促進皮膚傷口癒合。最後本篇的結果指出,蘋果多酚具有促進卵巢切除母鼠皮膚傷口癒合的良好效果。

The decrease of estrogen in menopause is an important factor to cause skin aging in women. It would also decelerate the rate of wound healing (WH). Decrease of estrogen could increase oxide metabolism and oxidative stress. In previous studies, many researchers focused on this issue and tried to find the ways of promoting wound healing in chronic diseases or aging. Apple polyphenols (APs) are proved to possess a good antioxidative effect, and it is beneficial for human. In this study, we address an issue to explore the effect of APs in promoting wound healing in the ovariectomic (OVX) rat. The rats were ovariectomized and convalesce for four weeks. The uniform burn wound on the rat back skin is artificial made in this study. The APs were mixed with base cream to treat on the wound via dressing for ten days. In histochemical stain, adipocyte under the skin wound increase, skin lesion appears and the hair follicle becomes rare and irregular. After treating with apple polyphenols for 10 days, the adipocyte showed decrease, skin repairs, and hair follicle recovers to line regularly, it shows a re-epidermalization and promote wound healing significantly as comparing to the control. In cell exam, this study using dermal layer of skin fibroblasts explore mechanisms for wound healing. After treatment of apple polyphenols, the migration protein p-FAK and MMP-2 was increased. Our present results show the apple polyphenols possess a good effect in wound healing under menopause.
