肉桂萃取物誘導口腔癌細胞凋亡之機制探討

摘要


肉桂是一種重要且很容易取得的傳統中藥材及香料。近年的研究指出,肉桂萃取物(Cinnamomum cassia extracts; CCE)含有多種活性物質,具有抗病毒、抗發炎、抗糖尿病、抗抑鬱、抗骨質疏鬆和抗癌效果,包括誘導癌細胞凋亡、抑制轉移等。然而肉桂萃取物對於口腔癌細胞的影響,尚待更多的探討。在本篇研究中,我們證實了肉桂萃取物會誘發SASVO3細胞凋亡反應,隨著肉桂萃取物濃度增加,對於細胞的毒殺性也隨之增加,明顯造成SASVO3細胞存活率下降、增殖受到抑制、細胞週期分佈sub-G1 phase比例增加、粒線體膜電位下降、細胞核染色質濃縮、Caspase‐3活化、Bcl-2蛋白表現量減少。另外由Annexin V及PI染色結果直接說明肉桂萃取物確實造成SASVO3細胞的凋亡。將來肉桂萃取物或許可應用在癌症化學預防及癌症輔助治療,幫助眾多的癌症患者。

Cinnamomum cassia is an important traditional Chinese medicine, and also used as spices. Recent studies have indicated that Cinnamomum cassia extracts (CCE) contains many kinds of bioactive substances that have antiviral, antidiabetic, anti-inflammatory, antiosteoporotic, antidepressive and anticancer effects, which involves the induction of apoptosis and inhibition of metastasis. However, the effects of CCE on human oral cancer need more studies. In this study, we demonstrated that CCE has cytotoxic effect toward a highly malignant oral cancer cell line, SASVO3. CCE reduced the viability, colony formation, mitochondrial membrane potential, and Bcl-2 protein of human oral cancer cells. Furthermore, CCE was shown to increase the percentage of cells in sub-G1 phase and induced the activation of Caspase-3 and the condensation of chromatin. In addition, the results of Annexin V/ PI staining indicated that CCE caused the apoptosis of SASVO3 cells. In the future, CCE may be used in cancer chemoprevention and adjuvant therapy.
