利用毛細管電泳分析半枝蓮的抗癌成分

The Separation and Quantification of Anti-carcinogenic Ingredients in Scutellaria Rivularis Wall by Capillary Electrophoresis

摘要


本研究是以毛細管電泳(Capillary Electrophoresis,CE)分離與定量半枝蓮(Scutellaria Barbate D.Don.)中具有抗癌作用的芹菜素(Apigenin)、黃芩甙(Baicalin)、粗毛豚草素(hispidulin)、木犀草素(luteolin)、柚皮素(Naringenin)、漢黃芩素(wogonin)與原兒茶酸(protocatechuic acid)等七種成分,同時也探討不同變數對各成分在遷移行為上的影響。 分析結果以微胞電動層析( Micellar Electrokinetic Capillary Chromatography, MEKC)中使用pH10.0且添加有20 mM SDS以及5%(v/v) isopropanol的15mM硼酸鹽緩衝溶液成功地將Apigenin、Baicalin、Hispudilin、Luteolin等四種類黃酮物質與半支蓮其他成分分離;使用含有20mM SDS、pH 8.75的25mM硼酸鹽緩衝溶液能有效分離出Wogonin、Luteolin、Apigenin等三種類黃酮物質;至於Naringenin與多酚酸Protocatechuic acid則是在毛細管區帶電泳(Capillary zone electrophoresis,CZE)的模式下即可達到與其它成分分離的目的。 在最佳化之分離條件下,各成分之標準添加曲線均呈現良好之線性關係,線性迴歸模式適切度之判定係數R2值皆為0.997以上。同日間(intra-day)與異日間(inter-day)試驗之相對標準偏差皆小於3%,回收率試驗顯示本分析方法有良好的再現性及準確性,可作為半枝蓮定量的分析條件。 使用索式萃取法,以50%(v/v)乙醇水溶液為溶劑,萃取半枝蓮樣品五小時,可求得5g的半枝蓮樣品中分別含有1627 ppm的Apigenin、66 ppm的Baicalin、213 ppm的Hispidulin、715 ppm的Luteolin、44ppm的Wogonin、59 ppm的Naringenin 以及62 ppm的Protocatechuic acid。

An analytical method was developed to separate and quantify the anti-carcinogenic ingredients, Apigenin, Baicalin, Hispidulin, Luteolin, Naringenin, Wogonin and Protocatechuic acid in the Scutellaria rivularis Wall by capillary electrophoresis. The relationships between the migration behavior and various variables were also studied. Apigenin, Baicalin, Hispudilin and Luteolin have been successfully separated from Scutellaria rivularis Wall by micellar electrokinetic capillary chromatography (MEKC) with a pH 10.0 buffer containing 15mM Borate, 20mM SDS and 5%(v/v) isopropanol. Wogonin, Luteolin and Apigenin can be separated with a pH 8.75 buffer containing 25mM Borate and 20mM SDS.Naringenin and Protocatechuic acid can be separated at a pH 9.3 buffer containing 30mM Borate by capillary zone electrophoresis (CZE). Under optimizing conditions, all standard addition plots of the instrument response versus added volume were linear over the entire ranges tested with coefficient of determination (R2 value) for the fitness of the linear regression larger than 0.997.The relative standard deviations (R.S.D.) of peak areas for both intra-day and inter-day tests were less than 3%. The developed analytical method was proved to be reproducible and precise. Scutellaria rivularis Wall, which was extracted by Soxhlet extraction with 50%(v/v) ethanol for 5 hour, contains 1627 ppm of Apigenin, 66 ppm of Baicalin, 213 ppm of Hispidulin, 715 ppm of Luteolin, 44 ppm of Wogonin, 59 ppm of Naringenin and 62 ppm of Protocatechuic acid.
