藥物與剪應力對人類主動脈內皮細胞的影響

摘要


本論文主要是探討加入藥物後以及不同剪應力下對人類主動脈血管內皮細胞的影響,本研究首先先架設一簡易體外細胞培養系統,將血管內皮細胞培養在波片上,實際培養於流體腔中,並給予符合人體血管的剪應力,結果顯示本系統設計與實驗是可行的,再給予不同的剪應力後,細胞型態上從橢圓形改變成長條形,這型態的改變證實經過本系統架設的流體腔在給予一定時間後細胞的型態會隨著水流方向改變,而改變後的型態也符合真實人體在有血流的情況下內皮細胞的型態。本論文使用的藥物為Cilostazol主要探討的是加入藥物後對細胞的影響,首先先架設上述的培養系統,在培養液中加入定量的藥物濃度,在固定時間即剪應力後發現,經過計算後細胞數目有少許的增加,而加入藥物跟未加入藥物的細胞比較下,加入藥物後的細胞較圓,也證實經過本系統架設的流體腔可以初步的模擬藥物進入人體對細胞的影響。

In this paper, the main subject is to discuss the influence of medicating Cilostazol medical and regulating the different shear stress to human endothelial cell. First step is to set up a simple cell culture system in vitro. Then the endothelial cells were cultured on plates, and modulated the shear stress as human environment for the chamber. As the results show that endothelial cells have changed its morphology as the flow direction that is investigated as factual in human environment. As to confirm the quantitative concentration of medical that affect the endothelial cells, the shear stress and the periods of time is constant. Obviously, the density of endothelial cell has a tendency toward to increase. In comparison with non medicated, adding Cilostazol medicine is more stable to be its origin morphology. However, the medicine effect and the flow chamber system to human endothelial cells is investigated.
