細胞外基質被證明與細胞的遷移、生長與分化有關係。最近,在軟骨修復與再生上,越來越多的細胞外基質(ECM)被製作成支架來提供獨特的生長環境。 本研究的目的為發展可以在避免免疫排斥反應下,又能保留其天然的軟骨成分的豬來源去細胞軟骨基質(ACM),應用於組織工程上。 結果顯示幾乎所有DNA都被大量的去除而醣胺多醣(GAG)則能成功保留45% ~ 74% 的含量。而在組織免疫染色上,沒有alpha-gal被觀察到。在體外培養實驗發現,去細胞軟骨基質(ACM)能與生長因子協同性地增強骨髓間葉幹細胞軟骨分化的效果。總結為去細胞基質有能在組織之工程中當作具有利於軟骨分化的功能性支架。
The extracellular matrix (ECM) has been shown to provide cues that affect cell migration, proliferation, and differentiation. Recently, more and more extracellular matrix (ECM)-derived scaffolds have been examined and fabricated to offer a specific environment for cartilage repair or regeneration. The aim of this study is to develop porcine Acellular Cartilage Matrix (ACM) that both avoids immune-mediated rejection and retains natural composition for tissue engineering. The result showed that the processes completely removed DNA of ACM and retained 45%~74% glycosaminoglycan (GAG) content of ACM. There was no evidence of the expression of the alpha-gal epitope. The findings of gene expression suggested that the combination of ACM powders and growth factors showed a synergistic effect with regard to dominating chondrogenesis of bone marrow stem cell (BMSC) in vitro. In summary, ACM powders could serve as a functional scaffold that benefited the chondrogenesis of BMSC for cartilage tissue engineering.
