載體對肝細胞行為之影響

The influence of driver on the hepatocyte culture

摘要


肝臟是人體重要器官之一,每年仍有許多病患正等待健康肝臟移植。他們可以暫時取代肝細胞功能。 細胞在不同微載體下培養生長,會影響細胞的型態和功能,可測特定hepG2功能,選出較適當肝細胞生長環境。肝細胞屬於易聚集型態,且此特性能保持穩定肝細胞特有功能和較高的代謝功能,如白蛋白分泌、尿素合成、代謝與解毒等功能。所以藉助此特性,設計不同類型的細胞載體去促使細胞自然聚集成球狀,誘導細胞產生極化使之分化出肝細胞特有肝功能。 此研究我們採用4種不同材料,三維立體空間的聚乙烯醇混合幾丁聚醣之奈米纖維薄膜、聚乳酸混和幾丁聚醣奈米纖維、表面有柱狀的聚二甲基矽氧烷 (PDMS)材料,與二維平面材料的24孔盤作比較,個別觀察它們對肝細胞生長的影響。 實驗結果顯示三維結構材料確實可以使肝細胞自然聚集成立體結構,尿素合成與代謝也比二維環境來的高。

The liver is a vital organs in the animal . Every year, many patients waiting for replacement of health liver,so that develop to different liver cell culture devices, consisting of functional liver cells and artificaial material, they can be a temporary organ replacement. In different driver to culture hepG2, it will influence their hepG2 configuration and function. It can assay hepatocyte’s function, to chose suitability growth environment. Hepatocyte’s property is aggregation spontaneity and it will maintence of specific liver’s function and high metabolic activity. ex: albumin of secretion,urea synthesis,metabolic activity. Apply this property , to design different culture environment to improve cell aggregate spontaneity to induce membrane proteins into a polarized distribution . This research ,we study 2 kinds of different type of cell culture microcarrier to induce hepatocyte aggregate . This result, we can observed 3D environment include of pva/cs nanofibrous membrane、3D PDMS with pillars membrane and 24well plate of 2D plane culture , result is 3D environment it can improve the cell to aggregate, and urea synthetic’s activity is higher than 2D environment.
