硫複鐵礦製程與癌症溫熱療法之應用

摘要


溫熱療法應用於惡性腫瘤治療已有數十年歷史,利用惡性腫瘤與正常組織間血液交換量低而造成散熱性不佳的特點,將惡性腫瘤所在區域溫度提升至43到46℃,使得惡性腫瘤無法快速散熱而凋亡。而感應式溫熱療法是溫熱療法應用的一種,是將磁性粒子注入人體,利用磁性材料在外加交變磁場下的磁滯損失作用進而產生熱,能有效率的集中並提升治療區域的溫度。此法改進了傳統溫熱療法只能治療淺層且發熱區域不集中的缺點,而且若將磁粒粒子當成熱種子,配合適當的表面修飾,能直接被癌細胞攝取進入細胞內部,更能有效直接使癌細胞凋亡。本實驗利用鐵硫化合物中具有磁性的中間相化合物硫複鐵礦為主要研究材料,研究並嘗試以低成本、快速的方式製作出磁性硫複鐵礦顆粒。實驗中發現,利用共沉法方式製作硫複鐵礦時,以XRD觀察不同時間不同pH值合成鐵硫化合物產物情形。本實驗以共沉法在反應溶液於pH=3,10分鐘時製備出現硫複鐵礦。利用超導量子干涉元件檢測硫複鐵礦磁滯迴圈並計算其釋放的能量為每毫克每秒0.02273卡。在頻率1.1×106MHz感應式磁場作用下,低濃度的混合溶液到達43℃花費8分鐘,隨著濃度增加,到達43℃所花費的時間更短。而根據比吸收率的計算下,在單位時間內每克水溶液吸收了2.1607瓦特。而進行WST-I以及LDH基本細胞毒性測試時發現硫複鐵礦在濃度為1mg/ml的情形下對細胞的傷害性最小。同時經過In Vitro的實驗證明,硫複鐵礦在外加交變磁場的作用下確實能夠殺死癌細胞。

Hyperthermia is novel therapy for malignancies in recent years. Malignant tumors are exposed in 43~46℃ successively for 30 minutes and malignant tumor are damaged or killed in this situation. Conventional hyperthermia was only applied shallow regions and heat was not focused accurately. In order to solve these problems, some scientists use magnetic particles could be used seed to target cancer cells which were taken by receptor medium endocytosis and were produced heat with extra AC magnetic field by hysteresis which was called inducted hyperthermia. Greigite is Fe-S complex and has magnetic property. It is only synthesized in the solution. This experiment is synthesized greigite by precipitation in different pH value and reactive times. It is checked phase of greigite with X-ray diffraction and found appropriate parameter in pH=3 and 10 minutes. Greigite is calculated the energy by area of hysteresis loop in 10000 Oe in SQUID and it is released 0.02273 . We used different concentrate of greigite to carry out heating with Inductive Heater Powder Cube, it is arisen to 43℃ at 8 mins in 1mg/ml in 1.1×106MHz. In LDH and WST-1 are revealed low toxicity in concentrate of 1mg/ml of greigite. For in vitro experiments, greigite are arisen temperature certainty by extra magnetic field to kill cancer cells.
