LabCD在洋蔥DNA萃取之應用

摘要


微流體技術在生物晶片中快速發展,其微量總體分析系統(Micro Total Analysis System)便是將生醫檢測上取樣、樣本傳輸、混合、分離及偵測等功能。 本論文發展四種碟片,分別為(一)紅、黑墨水混合碟片(二)紅、黑、藍墨水混合碟片(三)洋蔥DNA碟片使用紅、黑、藍墨水進行混合之模擬碟片(四)洋蔥DNA碟片使用實際液體碟片,藉由NTFL實驗室所推導出臨界轉速公式,經過基礎轉速與應用碟片兩者實驗比對後,預測突破閥門的轉速誤差在10%內,相當精確且實用性,藉由NTFL可攜式光碟機,使馬達正、反轉作混合效果,吾人稱為振盪混合法,本文碟片的混合效果相對誤差分別為14%、17%、30%。 最後吾人成功開發出洋蔥DNA碟片使用實際液體碟片,且經紫外光測量和DNA電泳加以來驗証有DNA存在。希望未來能結合生醫知識多方之應用,擴大LabCD之應用面。

Micro-fluid techniques are developed rapidly in the bio-chip area. The Micro Total Analysis System provides the capabilities in sampling, transporting, mixing, separating, and sensing function. This thesis develops four kinds of bio-discs, first red black ink mixing the disc, second red black blue ink mixing the disc, third onion DNA disc is used red black blue ink mixing the simulation disc, fourth onion DNA disc uses the real liquid disc.The critical rotational speed formulae derived from NTFL laboratory, basic rotational speed and application disc two experiments, predict that it is in 10% to break through the error of rotational speed of the valve.NTFL Portable CD-ROM,the motor cw/ccw make mix, we called shaking mixing method, that bio-discs mixing effect relative error 14%, 17%, 30%. Finally, we succeed in developing onion DNA disc uses the real liquid disc, Proved DNA exists by UV Spectrophotometer and electrophoresis. We hope this research can be combined with more biomedical applications, and increase the applicability of LabCD.
