探討FUCA1(α-L-Fucosidase) 在乳癌中所扮演的角色

摘要


本研究主要目的是在探討乳癌與FUCA1(α-L-Fucosidase)的關聯性。過去的臨床文獻指出透過利用多種不同篩選技術找尋與癌症相關的基因,發現FUCA1(α-L-Fucosidase)跟乳癌是具有相關性的,但文獻僅專注在篩選可作為診斷用的標誌,而沒有深入探討FUCA1(α-L-Fucosidase)在乳癌中扮演著如何的角色。 本研究在臨床上,利用乳癌病人的檢體,首先先利用Real time PCR(RT-PCR)觀察FUCA1在乳癌細胞系的表現,再以Real time PCR來觀察FUCA1在臨床乳癌病人組織的表現量,更進一步利用免疫組織化學染色(IHC)的方法,確認其蛋白表現分佈,結果皆發現FUCA1在 II 乳癌細胞系及乳癌病人的腫瘤組織表現平均比正常組織來的高36倍並具有顯著意義。另一方面,利用實驗室現有的乳癌細胞株,探討其調控作用機制。 近年來有研究指出,在肝癌中 FUCA1 能扮演一個腫瘤標誌的角色,而 FUCA1 在腫瘤中所扮演的角色仍然不明確,為了探討這樣的現象是否也會發生在乳癌上面,透過FUCA1抑制劑Ethambutol和FUCA1si-RNA分別送入乳癌細胞當中,透過細胞存活率測定及生長曲線實驗,觀察其乳癌細胞生長及爬行的情形,由結果發現FUCA1具有抑制乳癌細胞生長及的能力以及侵略性。 在過去的研究認為癌症是醣質化過程中失去規則性而紊亂的現象所導致,以導致細胞間的辨識系統失控,最後變成侵略性的轉移。 本研究證實FUCA1在乳癌細胞中所扮演的角色能影響乳癌細胞的增生以及增加乳癌細胞的移行能力,並首次發現FUCA1的表現與下游蛋白FUT6具有相關。

The purpose of this study was to explore the correlation between breast cancer and FUCA1(α-L-fucosidase). The clinical literature indicates that in the past to find a biomarker for cancer by several screening techniques revealed that FUCA1 (α-L-fucosidase) was correlating with breast cancer. But those studies only focus on finding a potential biomarker, instead of figuring out the role of FUCA1 (α-L-fucosidase) in breast cancers. IV In this study, we used clinical breast cancer tissue to detect the expression of FUCA1 (α-L-fucosidase). From Real-time PCR data, we observed that FUCA1 (α-L-fucosidase) gene level in breast cancer tissues were higher than in normal tissues. Next, we use Immuno-histochemistry (IHC) staining to confirm the protein expression and distribution of FUCA1. The results are found FUCA1 in breast cancer cell lines and breast cancer tumor tissue than normal tissue to the average performance of the high. On the other hand, we use of existing laboratory breast cancer cell lines to studies the regulatory mechanism. In recent years, research indicates that in liver FUCA1 can play a role in tumor markers, and FUCA1 in the role of the tumor is still not clear, in order to explore whether this phenomenon also occurs in breast cancer above, through FUCA1 inhibitors Ethambutol and FUCA1 si-RNA, respectively, into breast cancer cells which, through the determination of cell survival and growth curve experiments, observe the breast cancer cell growth and crawling the case, the results showed inhibit FUCA1 can effect the ability of cell V growth and maigration in breast cancer `` In the past studies suggest that cancer is the loss of the rules of carbohydrate in the process ,the disorder caused by the phenomenon, leading to cell-cell recognition system out of control, eventually become aggressive metastasis. This study demonstrated that FUCA1 role may increase the free-from fucose to provide its downstream protein utilization of fucose in order to facilitate increased cellular glucose qualitative phenomenon in breast cancer cells.
