中文摘要 論文名稱:探討山竹中純化之天然化合物Garcinol對乳癌細胞株內 Cyclin D3 表現之影響 研究所名稱:台北醫學大學醫學科學研究所醫學檢驗暨生物技術組 研究生姓名:謝承達 畢業時間:九十八學年度 第二學期 指導老師:何元順 博士 台北醫學大學醫學檢驗暨生物技術研究所教 授 本研究主要是探討乳癌中過量表現的Cyclin D3和山竹粹取物:Garcinol之間的關聯性。在流行病學的研究中可以看出,台灣婦女罹患乳癌的比例逐年升高,可能由於環境的逐漸西化加上生活飲食的改變。為了有效的改善和預防乳癌的發生率,從各種天然粹取物研究是否有既安全又有效的方法。山竹是富含營養的水果,而其粹取物Garcinol在In vitro的實驗中可以有效的抑制惡性的乳癌細胞MDA-MB-231生長,而其抑制的途徑是藉由抑制細胞生長週期中的Cyclin D Family的Cyclin D3表現量,使細胞週期停滯。另一方面,在乳癌病患的檢體中發現,除了已報導過的HER2等基因外,Cyclin D3的含量也高於正常的細胞。從Luciferase assay中發現Garcinol可以有效降低Cyclin D3 promoter的AP-1 binding site的作用。因此結合兩者的結果,希望可以藉由研究Garcinol來改善乳癌病患的情況,或者可以提早預防乳癌的發生率。

Abstract Title of Thesis: Cyclin D3 is down-regulated by Garcinol in MDA-MB 231 breast cancer cells Institule: Graduate Institute of Biomedical Technology, Taipei, Medical University Author: Chang-TA Hsieh Thesis advised by: Yuan-soon Ho, Ph.D. (Professor, School of Medical Laboratory Science and Biotechnology,College of Medicine, Taipei Medical University) The purpose of this study is to figure out the correlation between over expression of Cyclin D3 in breast cancer and the components of Mangosteen:Garcinol. In the recently study of epidemiology, the higher rate getting breast cancer of women in Taiwan is serious year by year. The reason maybe is the change of lifestyle and westernization. For cure or prevention of breast cancer effectively, we try to find the components is safely and healthy from nature components. Mangosteen is a kind of very nutritional fruit. Its extraction, Garcinol, is effectively suppressing the breast tumor, MDA-MB-231, growth in vitro. The pathway is via downregulation of Cyclin D3 expression, which is an important protein regulating cell cycle. The other way, we found the cyclin D3 over expression in clinical breast cancer tumor tissue than in normal tissue. In Luciferase assay, we found that Garcinol can suppress transfection factor binding on the AP-1 site of Cyclin D3 promoter. Because of these results, we research into Garcinol for cure or prevention in breast cancer.
