摘要


我們二維電泳技術觀察大腸桿菌再不同抗訊藥物處理下產生的生理反應.我們建立了關於不種類抗細菌藥物的二維電泳資料庫,在pI 3到10範圍之內.不同藥物處理之下會產生不同的蛋白質圖譜.藉著較新的有抗細菌活性的藥物與已知機制的抗細菌藥物的圖譜,可以初步的分類新藥維哪一種機制.在本篇當中可以將新藥(755a)做一個初步的分類.這樣的蛋白質技術提供一個新藥開發一個新的觀點,也提供一個有用的工具加速新藥的開發.(專利申請中,暫不公開)

We have used proteomic technology to elucidate the complex cellular responses of Escherichia coli to antimicrobial compounds belonging to classical antibiotic classes. We established a two dimensional comprehensive database of total proteins with pIs covering a range of 3 to 10 during treatment with antibiotics known to cause cell damage. Each antibiotic showed a complex and characteristic protein expression profile suggesting that new compounds with unknown mechanisms of action may be classified. Indeed, one such substance (755a). These results suggest that this technique gives new insights into the bacterial response toward classical antibiotics and hints at modes of action of novel compounds. Such a method was useful in the process of antibiotic drug discovery (專利申請中,暫不公開).
