沉香葉的化學成分及傷口癒合活性之研究

摘要


近年來一些新的研究顯示出,在傷口癒合期間,適當的促進MMP-9和MMP-2的活性有助於減少疤痕的形成。在傷口癒合活性的生物活性篩選中,發現沉香葉的粗萃物具有顯著促進proMMP-9和MMP-2的活性。因此,為了找出具有傷口癒合活性的化合物,本研究將沉香葉部的甲醇萃取物,進行了分離純化及鑑定。目前從沉香葉中,已分離出28個化合物,這28個化合物包含脂肪酸類、色酮類、黃酮類、木質素、醣苷類、固醇類以及萜類。這些化合物的結構,利用了各種不同的光譜儀器鑑定,包含IR, UV, MS, 1D及2D NMR;另外,MMP的活性則是以Zymography的方法來測定。結果發現,化合物AQ20具有顯著促進proMMP-9和MMP-2的活性(EC50 = 21.95, 15.82 μM)。本研究另外將五個AQ20的類似物也進行的活性測定,分別命名為20A~E,但最後結果其對於MMP-9和MMP-2並無顯著差異。目前的研究顯示出,化合物AQ20具有幫助傷口癒合的潛力且不具有細胞毒性,但還需要更進一步的研究來證實,在未來需進一步探討AQ20的SAR,並期望本研究在對傷口癒合上能夠發揮一定的效用。

In recent years, new research shows that appropriately promote proMMP-9 and MMP-2 activities in specific phase of wound healing, which could help to reduce scar formation. Base on bioassay guided of wound healing activity, we found that the crude extract from leaves of Aquilaria sinensis showed significant promoting activities for proMMP-9 and MMP-2. Therefore, the aim to find active wound healing component(s), we isolated and purified the MeOH extract from the leaves of A. sinensis. We have been isolated 28 compounds from A. sinensis. Now, twenty compounds has been identified including aliphatic, chromones, flavonoids, ligands, glycosides, steroids, and terpenoids. All of the structures were identified by extensive spectroscopic analyses, including IR, UV, MS, 1D and 2D NMR. We measured MMP activity by Zymography method. As a result, the compound AQ20 has significant stimulating activities for proMMP-9 and MMP-2(P<0.001) and no cytotoxicity, with EC50 value of 21.95 and 15.82 μM. Base on the result, we measured five analogues of AQ20 which named 20A-20E. The results shows that 20A-20E no significant stimulating activities. Eventually, the present study explored the wound healing potential of compound AQ20. In future work we will test it in vivo study and investigate the SAR (Structure-Activity Relationships) of AQ20 and its analogues.
