背景:葡萄為富含多酚類物質的水果,對於改善或提升老年認知功能有顯著的效益。然而,葡萄攝取對於改善高血糖相關的認知功能受損的機制在過去研究上仍非常有限。目的:探討葡萄攝取對於長期高果糖高脂飲食老年大鼠,對於空間學習與記憶表現以及分析腦部皮質與海馬迴相關神經退化蛋白表現量的影響。方法:研究選用6週齡雄性Wistar大鼠,以高果糖高脂飲食餵養至66週齡後,介入metformin、低劑量 (3% w/w)與高劑量 (6% w/w)葡萄粉飲食12週。分別於大鼠52和78週齡以莫氏水迷宮分析大鼠的空間學習與記憶表現,犧牲取得大腦組織以西方點墨法分析腦部皮質與海馬迴相關蛋白的表現量。結果:葡萄攝取對於長期高果糖高脂老年大鼠可顯著提升空間學習與記憶表現,腦部皮質與海馬迴Aβ和RAGE蛋白表現量降低,Nrf2和BDNF蛋白表現量上升。結論:大鼠攝取含6%葡萄粉的飲食12週,可改善長期餵養高果糖高脂飲食老年模式下空間學習與記憶受損。因此有潛力應用於飲食改善老年認知功能受損。

Background: Grape, which is rich in polyphenols, has been suggested to improve cognitive function in aged people. However, the mechanisms that grape consumption reverse hyperglycemia-related cognitive dysfunction remains unclear. Purpose: To investigate the effects of grape consumption on spatial learning, memory performance and neurodegeneration-related protein expressions in aged rats fed high fructose high fat (HFHF)diets. Methods: 6 weeks old Wistar rats were fed HFHF diets to 66 weeks old and started the intervention diets. The intervention diets were treated metformin, low and high dose grape powder for 12 weeks. The spatial learning and memory performance was assessed by Morris water maze. Cortex and hippocampus protein expressions were assessed by Western blotting. Results: Grape consumption significantly increased spatial learning and memory performance, reduced Aβ and RAGE protein expressions, and up-regulated Nrf2 and BDNF protein expressions of cerebral cortex and hippocampus in aged rats fed HFHF diets. Conclusion: Diets contain 6% grape powder consumption for 12 weeks ameliorated spatial learning and memory performance impairment in aged rats fed HFHF diets.
