Abstract The major purpose of this research is to investigate the optimization of producing polyhydroxyalkanoates (PHAs). In order to rise the product, the medium and parameter of fermentation will be changed in this research. First of all, using LB and Thioglycolate medium to produce PHAs in the condition of 30℃、200rpm. For Thioglycolate medium, the result 2.22g/L bacterium dried weight, 9.83% PHB and 0.7% HV can be got. However, for LB4 medium (1.5g Tryptone、0.5g Yeast extract、1.0g NaCl) the result 2.55g/L bacterium dried weight and 19.82% PHB can be got. For the fermentation parameter, the optima conditions are 30℃ and pH7.0. Furthermore, the olive oil will be used as carbon source heated in 250℃ for 10 minutes will get the best result 2.93g/L dried weight and 30.19% PHB content. Taking this carbon source condition into Thioglycolate medium, both dried weight and PHB content will not increase obviously but the HV content will be up to 4.99%. Otherwise, as being the condition C/N = 2/1(0.5 ﹪Olive oil : 0.25﹪NH4Cl) the best consequent can be observed 4.49g /L dried and 33.91 PHB content.
