摘要


選擇性剪接(alternative splicing) 機制可使同一個基因產生不同的訊息核醣核苷酸(mRNA),進而轉譯為不同的蛋白質產物。本研究以全長cDNA序列對水稻與阿拉伯芥兩植物基因體作定位與分群,藉由觀察各群外顯子(exon)或內含子(intron)重疊的情況以偵測各種類型的選擇性剪接。我們考慮AA、AD、IR、ES、A3與A5類型的選擇性剪接,在阿拉伯芥分析的16,604個基因當中,有4,036個基因(佔24.3%)具有選擇性剪接現象,一共偵測出12,818筆選擇性剪接事件。在水稻方面,我們分析的21,319個基因當中,偵測出3,991個(佔18.7%)有選擇性剪接的基因,一共找出了17,632筆選擇性剪接事件。水稻發生選擇性剪接的基因當中,有33.6%的基因發生IR型態的選擇性剪接,而發生ES的基因則佔了5.1%;在阿拉伯芥中發生IR型態的選擇性剪接的基因比例約為9.3%,而發生ES的基因所佔比例為2.9%,我們發現ES事件在兩物種中發生的比例皆是最低的。

Alternative splicing is a mechanism that can make one gene to produce various different messenger RNA (m-RNA), and then can generate various different protein products. We detected various different types of alternative splicing events based on clustering full-length cDNA sequences on Arabidopsis thaliana and Oryza sativa genomes, and discovering of several exons or introns in the same cluster. We consider AA, AD, IR, ES, A3, A5 type of alternative splicing events. We find there are 4,036 (24.3%) genes that have alternative splicing out of total 16,604 genes in Arabidopsis thaliana, and 12,828 alternative splicing events have been detected in total. In Oryza sativa, we find there are 3,991(18.7%) genes that have alternative splicing out of total 21,319 genes; meanwhile, we detected total 17,632 alternative splicing events. Among Oryza sativa genes that have been observed for alternative splicing, 33.6% genes occurs IR event, while 5.1% genes occurs ES event. On the other hand, IR event is present in 9.3% of Arabidopsis thaliana genes, and ES event occurs in 2.9%.
