製作環境控制掃描式共焦顯微鏡應用於單分子光譜檢測

摘要


本論文之研究是製作一台精巧、低成本、功能強大的共焦顯微鏡系統,並用LabVIEW撰寫該顯微鏡之控制程式與數據分析,做單分子的區域掃描。一個標準的單分子實驗需要樣本的準備與光柵的掃描,並且尋找單分子分佈的位置,接著利用時間追跡(Time trace)去了解光致螢光的改變情形。我們利用這台顯微鏡去探索共軛高分子對環境的光物理影響,並且藉著單分子的生命期(Lifetime)了解溼度與氧氣,是否會造成單分子的快速衰退的行為,與探索共軛高分子螢光熄滅的物理變化。

We propose to set up a compact confocal microscope and write controlling programs of confocal microscope by LabVIEW. A standard single molecule experiment involves do raster scanning and prepared sample to find a single molecule, or position a single molecule in the focus and observing changes in photoluminescence with time. In addition, new algorithm is developed to allow high speed single molecule photon correlation spectroscopy. We hope to utilize this confocal microscope to explore the effect of environment on the photo-physics of luminescent conjugated polymers answering whether humidity or oxygen is more detrimental to polymer lifetime and exploring the fluorescence quenching mechanism in conjugated polymers.
