本研究利用幾丁聚醣在生醫材料方面具有生物相容性、生物活性、生物可分解性及植入體內可降低排斥現象和無毒性的特性加以應用。將chitosan與gelatin共溶於醋酸溶液中,Sodium hyaluronate與chondroitin-6-sulfate共溶解於二次水中,然後chitosan/gelatin溶液與 HA/C6S 溶液在室溫下攪拌使之混合。再用甘油-2-磷酸二鈉水溶液滴入chitosan/gelatin/HA/C6S溶液直到pH值達到7.4為止,製作chitosan/gelatin/HA/C6S hydrogel之軟骨組織工程細胞載體。並且利用流變儀、以及各項生物相容性測試、電子顯微鏡觀察水膠以及水膠中之軟骨細胞、切片與染色,探討chitosan/gelatin/HA/C6S hydrogel之各項性質。再進一步研究添加HA/C6S 與否對chitosan/gelatin solution 作為細胞載體的差異,結果均為軟骨組織的良好細胞載體,並且添加HA/C6S可提高細胞的活性與細胞的存活度。
Chitosan with the characteristics of biocompatibility, biodegradability, non-cytotoxicity and low-immunogenic property is applied in biomedical fields widely. Thermal-sensitive chitosan hydrogel, which would transfer from liquid to gel in body temperature, is an alternative for cell carrier. In this study, gelatin was added into the chitosan hydrogel to improve the mechanical strength. In addition, sodium hyaluronate and chondroitin-6-sulfat were further added to form a chitosan/gelatin/HA/C6S hydrogel for cartilage tissue engineering. For the analyses of this developing hydrogel, rheological measurement, material-mediated cytotoxicity, cell viability, glycosaminoglycan contents, scanning electron microscopy and histologic examinations were performed. The purpose of study was to demonstrate the influence of the additions of HA/C6S to the chitosan hydrogel related to the chondrocytes.
