枯草桿菌ilvC的選殖並探討其對表面素生產之影響

摘要


表面素為枯草桿菌ATCC21332以非核醣體胜肽合成機制所合成的脂胜肽生物界面活性劑,結構為由七個胺基酸所組成的環狀胜肽鏈,以及ㄧ長碳鏈脂肪酸側鏈所組成。ilvC是枯草桿菌合成Valine、Leucine、Isoleucine 胺基酸所需的酵素,而這些胺基酸為表面素胜肽鏈的主要組成。將ilvC以穿梭質體送入枯草桿菌SWK569,觀察表面素的產量。卻發現表面素產量只有原先的十分之ㄧ,且於固態培養基上發現菌體的活性會快速消失,並且死亡。推測是因為大量表現ilvC,始得菌體的生長受到影響,進而影響表面素的產量。

Bacillus subtilis need ilvC the enzyme to product Valine, Leucine, Isoleucine, and thesis amino acid are the main to structure the peptide chain。Using shuttle vector ransforming ilvC to Bacillus subtilis SWK569,but the deci-product of SWK569 only。Finding the bereft activity of SWK569 would die on plate。Calculation that was because the over expression of ilvC to effect the physiology of SWK569, and effect the productivity of SWK569。
