臺灣固有種植物內生真菌Colletotrichum aotearoa及Phomopsis theicola二次代謝產物及生物活性之研究

摘要


Colletotrichum aotearoa 及Phomopsis theicola為台灣固有種植物之內生真菌。Colletotrichum aotearoa和Phomopsis theicola分別是從野牡丹科金石榴之葉部及樟科小梗木薑子之葉部所培養出來的内生真菌。其中Colletotrichum aotearoa是以液態發酵培養,Phomopsis theicola則是以固態培養而得的。 Colletotrichum aotearoa 之正丁醇萃取物以乙酸乙酯及水進行分配,接著將乙酸乙酯可溶層以管柱層析法進行分離,得到了四個新的isochorman glycoside化合物:colletobredins A–D (357–360),兩個新的isocoumarin類的化合物:colletomelleins A and B (361–362)及12個已知化合物,分別為ergosterol peroxide (98)、monopalmitolein (363)、palmitic acid (364)、3-indole acetic acid (365)、akodionine (366)、cyclo(L-Leu-L-Pro) (367)、brevianamide F (368)、n-butyl β-D-glucopyranoside (369)、4-hydroxybenzoic acid (370)、4-methoxyphenol (371)、9,11-dehydroergosterol peroxide (372)、regiolone (373)。 另外利用活性導向分劃法由Phomopsis theicola之乙酸乙酯可溶部進行分離,得到11個化合物,其中包含一個新的cytochalasin類的化合物;phomocytochalasin (374),五個已知化合物,分別為cytochalasin H (228)、cytochalasin N (230)、RKS-1778 (375)、dankasterone B (376)、cyclo(L-Ile-L-Leu) (377) 及五個待解化合物。上述各化合物皆由各種圖譜分析決定其結構。

Colletotrichum aotearoa and Phomopsis theicola were two endophytic fungi found in the Taiwanese endemic plants. The former was isolated from the leaves of an endemic plant, Bredia oldhamii (Melastomataceae), processed through liquid fermentation and the latter was isolated from the leaves of an endemic plant, Litsea hypophaea (Lauraceae), cultured by solid-state fermentation. Bioassay-guided fractionation of the ethyl acetate layer from C. aotearoa led to the isolation of four new isochorman glycoside, colletobredins A–D (357–360),two new isocoumarin, colletomelleins A and B (361–362), along with 12 known compounds, ergosterol peroxide (98)、monopalmitolein (363)、palmitic acid (364)、3-indole acetic acid (365)、akodionine (366)、cyclo(L-Leu-L-Pro) (367)、brevianamide F (368)、n-butyl β-D-glucopyranoside (369)、4-hydroxybenzoic acid (370)、4-methoxyphenol (371)、9,11-dehydroergosterol peroxide (372)、regiolone (373). Bioassay-guided fractionation of the ethyl acetate layer from P. theicola led to the isolation of one new compound, phomocytochalasin (374), along with five known compounds, cytochalasin H (228), cytochalasin N (230), RKS-1778 (375), dankasterone B (376), cyclo(L-Ile-L-Leu) (377) and five unsolved compounds. The structures of these compounds were elucidated by spectroscopic techniques.
