細胞性免疫反應影響不同品系鼷鼠對廣東住血線蟲感受性之研究

中文摘要 不同品系的鼷鼠對感染廣東住血線蟲(Angiostrongylus cantonensis)的感受性有很大的差異,為瞭解其原因,本研究對低感受性的C57BL/6J (B6) 及高感受性的BALB/cJ (BALB/c) 兩個品系的鼷鼠各感染50隻廣東住血線蟲第三期幼蟲(Ac L3),結果B6 鼷鼠感染後平均存活天數超過30天,顯著高於BALB/c鼷鼠的25.6±1.5天(Chi-Square, p<0.01)。B6鼷鼠感染30隻Ac L3第21天自腦部回收的蟲數開始顯著少於BALB/c鼷鼠(Student’s t test, p<0.01)。感染後第21天回收自B6鼷鼠所回收的雄蟲的長度開始顯著短於BALB/c鼷鼠所回收的雄蟲長度(Student’s t test, p<0.01);回收自B6鼷鼠的雌蟲長度在感染後7天即顯著短於BALB/c鼷鼠所回收的雌蟲長度(Student‘s t test, p<0.01)。兩品系鼷鼠感染後,末稍血液嗜伊紅性白血球百分比均沒有顯著差異(Student’s t test, p>0.05);而B6鼷鼠感染後第21天腦脊髓液中嗜伊紅性白血球百分比開始顯著高於BALB/c鼷鼠(Student’s t test, p<0.01)。兩品系鼷鼠脾臟細胞於體外分別與ConA、LPS、Ac L3抗原及Ac L5抗原共同培養,測定培養液內細胞激素的含量,大致而言,除了Ac L3刺激產生的IL-2以外,兩品系鼷鼠感染後至28天IFN-γ及IL-2比感染前的濃度低;感染後以Ac L3及Ac L5抗原刺激脾臟細胞,結果顯示B6感染後7天及14天IL-4的濃度顯著高於BALB/c (Student’s t test, p<0.05),而且感染後至28天期間B6的IL-5濃度均顯著高於BALB/c (Student’s t test, p<0.05)。因此推論BALB/c鼷鼠對廣東住血線蟲感染之感受性高於B6鼷鼠,可能與感染後所產生的調控嗜伊紅性白血球分化成熟的IL -5濃度有關。

The susceptibility to the infection of Angiostrongylus cantonensis was greatly different among mouse strains. To reconfirm the different susceptibility between C57BL/6J (B6) and BALB/cJ (BALB/c) to the infection, B6 and BALB/c mice were infected with A. cantonensis, and then cellular immune response were assayed in this study to elucidate the possible mechanism. The survival in days after infection with 50 L3 among B6 mice were significantly higher than those among BALB/c mice (p<0.01). The worm counts in brain ventricle became significantly less and the length of male worms became significantly shorter in B6 mice than that in BALB/c mice 21 days after infection with 30 L3 of A. cantonensis (p<0.01). However, the size of female worms from B6 mice 7 to 28 days after infection was significantly smaller than that from BALB/c mice. As to eosinophil counts, there was no significant difference between B6 and BALB/c mice among tailed blood, but, B6 mice showed significantly higher percentage than those of BALB/c mice among CSF 21 and 28 days after infection (p were all <0.01). Quantity of cytokines released in culture medium by spleen cells stimulated with mitogens included ConA, LPS, Ac L3 and Ac L5 were weekly carried out in mice until four weeks after infection. Generally, IL-2 and IFN-γin mice after infection were less than those of unifection except IL-2 in mice stimulated with Ac L3. IL-4 in B6 mice were significantly higher than those of BLAB/c mice only at day 7 and 14 day after infection (p were all <0.05). However, IL-5 in B6 mice from first weeks until the fourth week after infection were all significantly more than those of BALB/c mice (p were all <0.05). Thus, B6 mice produce more IL-5 after infection which stimulate the proliferation and maturation of eosinophils to against the worms by ADCC mechanism that lead B6 mice to lower susceptibility to the infection with A. cantonensis.
