以高效液相層析結合電化學檢測法進行台灣市售薑黃科學中藥產品之品質評估

Quality assessment of herbal extract products of Curcumae longae rhizoma sold in Taiwan by high-performance liquid chromatography with amperometric detection

摘要


薑黃是一種多酚類的天然化合物,它可以使用作為食品防腐劑,用於著色劑和調味品。薑黃在傳統醫學上被用於治療肝臟疾病,研究指出薑黃也具有抗氧化與抗發炎等活性。本研究目的為開發高效液相層析連接電化學偵測器的分析方法,應用於測定台灣市售薑黃科學中藥產品之分析與品質評估。分析方法所使用的管柱為Luna Phenyl-Hexyl Column (250 mm×4.6 mm, i.d. 5 um),移動相為乙腈、甲醇和0.01 mM Na2HPO4 (20:40:40, v/v/v, 用磷酸調整pH 3.0) 分離三種類薑黃素,流速設定為1.2 mL/min,注射體積量20 uL。同日間及異日間薑黃素分析結果,相對標準偏差 (RSD) 在同日為1.9-5.3 %,異日為4.0-5.3 %。高效液相層析分析方法確效結果,校正檢量線濃度範圍5-80 ug/mL 呈現線性關係 (r2>0.9987) 且準確度範圍為91.1-107.6 %。本分析方法建立及確效後,分析市售十種科學中藥,利用階層式分群法 (Hierarchical Clustering Analysis, HCA) 將十種科學中藥依照薑黃素的含量分群組。此方法可成功定量10家不同廠牌薑黃中薑黃素的含量並應用於台灣市售薑黃科學中藥的品質評估。

Curcumae longae is a polyphenolic naturally compounds and used as a food preservative, as a colouring and spice. Curcumae longae has been shown to possess anti-oxidant, anti-inflammatory, anti-amyloidogenic and anti-cancer activities. The aim of our study was developed a high performance liquid chromatography (HPLC) coupled with electrochemical detection method with rapid sample preparation and high sensitivity for the determination of herbal extract powder products of Curcumae longae rhizoma sold in Taiwan. Three curcuminoids were separated on a Luna phenyl-hexyl column (250 mm×4.6 mm, i.d. 5 um) by the mobile phase containing acetonitrile、methanol and 0.01 mM Na2HPO4 (20:40:40, v/v/v, pH 3.0 adjusted by orthophosphoric acid) with a flow rate of 1.2 mL/min and the injection volume was 20 uL. Intra-day and inter-day in curcumin analysis result the relative standard deviations (RSD) ranged from 1.9% to 5.3% for intra-day precision and from 4.0% to 5.3% for inter-day precision, respectively. According to HPLC was analysis method and validate result curcumin regression equation was linear (r2>0.9987) over the range of 5 to 80 ug/mL while the accuracy ranged from 91.1 to 107.6 %. This method of analysis was established and validated, the HCA classify 10 samples on the basis of the curcumin content in all samples. The result indicated that the developed method could successfully quantify the 10 samples of the curcumin components in Curcumae longae rhizoma and could be applied to detect and evaluate the quality of traditional Chinese medicines.
