本研究目的為探討無柄靈芝(Ganoderma resinaceum)萃取物在SD大鼠保肝模型與倉鼠降血脂模型中的活性。在SD大鼠保肝模型中,我們先用管餵方式給予SD大鼠無柄靈芝萃取物,一天兩次,持續三天,於第四天以D-Galactosamine (D-GalN) 400 mg/kg腹腔注射於SD大鼠,誘發急性肝炎,並於此前後各給予一次無柄靈芝萃取物,於第五天犧牲之。在倉鼠降血脂模型中,我們以高脂飼料(high-fat diet,HFD)餵食四週,誘導倉鼠之非酒精性脂肪肝,四週後犧牲。取犧牲之SD大鼠與倉鼠之肝臟與血液,進行肝功能與脂質檢測,並進行肝切片觀察。此外,我們也進行了MDA、SOD、GSH含量檢測。在保肝模型中,給予無柄靈芝萃取物的組別之AST與ALT的血清濃度明顯低於誘導組,且肝臟的portal area之發炎情形也較輕微。在降血脂模型中,給予無柄靈芝萃取物的組別之AST、ALT、總膽固醇(T-CHL)、低密度膽固醇(LDL)的血清濃度明顯低於誘導組,以及T-CHL/LDL之比值亦較低。無柄靈芝是傳統中藥-靈芝(G. lucidum)的同屬蕈類,我們的研究結果顯示無柄靈芝的口服給予對於急性肝炎與非酒精性脂肪肝有治療活性,因此無柄靈芝有潛力可以發展為保肝及降血脂製劑。
The aim of this study is to explore the hepatoprotective (in SD rats) and anti-hyperlipidemic (in hamsters) activities of Ganoderma resinaceum supplementation. Acute hepatic injury was induced with D-galactosamine (D-GalN; 400 mg/kg, i.p.) in SD rats. G. resinaceum was administered p.o. 3 days prior to D-GalN administration. In hyperlipidemic animal model, we developed HFD (high-fat diet)-induced non-alcoholic fatty liver disease (NAFLD) in hamsters. G. resinaceum was administered p.o. followed HFD administration. We made the liver functions and the lipids level examination and pathological examination in liver section. In addition, we made MDA, SOD and GSH content tests. In hepatoprotective activity model, serum GOT and GPT were decreased significantly in G. resinaceum administered group. Furthermore, inflammation was inhibited in portal area of liver. In hyperlipidemic animal model, serum GOT, GPT, lipids level of total cholesterol (T-CHL) and low-density cholesterol (LDL) were decrease in G. resinaceum administered group. The T-CHL/LDL ratio was also decreased. G. resinaceum is a kind of mushroom and similar to the famous traditional Chinese medicine: G. lucidum (Chinese name: LinZi). Our results indicated that administered G. resinaceum could be beneficial for treatment of acute hepatitis and NAFLD. G. resinaceum may have potential for being a hepatoprotective and anti-hyperlipidemic pharmaceutical.
