由台灣產麵包樹(Artocarpus communis)根部之成分研究中,繼續分離得3個新黃酮類衍生物,並分別命名為Biartocommunol (1)、Cycloisomulberrin (2)、Dihydroartomunin (3),化合物1-3之結構鑑定,依據光譜學之方法及相關文獻資料比對而得;同時化合物Cycloisomulberrin (2)顯示其對於A549人類肺癌細胞(human lung cancer cells)具有顯著之腫瘤細胞毒殺抑制活性。此外,將自麵包樹(Artocarpus communis Forst.)及馬來波羅蜜(Artocarpus elasticus)中所分離得到之三個新化合物Biartocommunol (1)、Cycloisomulberrin (2)、Dihydroartomunin (3)與九個已知化合物Artomunoisoxanthone (4)、 Artocommunol CC (5)、Artochamins B (6) and D (7) Dihydroartomunoxanthone (8)、Cycloartelastoxanthone (9)、 Cycloartobiloxanthone (10)、 Artelastoheterol (11)、 Artonol A (12),以瓊脂醣凝膠電泳定量分析方法探討化合物在氧化機制條件反應下對DNA切割之抗氧化活性。結果顯示Cycloartelastoxanthone (9)及Artelastoheterol (11)於氧化機制反應條件下,對於DNA具有顯著地增加Supercoiled form及減少Open-circular form 之作用。進一步於氧化機制反應條件下降低Cycloartelastoxanthone (9)之化合物濃度,結果顯示其仍具有顯著之抗氧化活性。
From the root of Artocarpus communis, three new prenylated flavonoids, biartocommunol (1), cycloisomulberrin (2), and dihydroartomunin (3), were further isolated. The structures of 1-3 were elucidated by spectroscopic methods and by comparison with data reported in the literature. The new compound 2, exhibited cytotoxic activity against the A549 human lung cancer cell line. The inhibitory effects of the above new compounds, 1-3 and nine known compounds, artomunoisoxanthone (4), artocommunol CC (5), artochamins B (6) and D (7), dihydroartomunoxanthone (8), cycloartelastoxanthone (9), cycloartobiloxanthone (10), artelastoheterol (11), and artonol A (12) isolated from A. elasticus on protective effect of DNA damage caused by superoxide anion (generated by XA/XO) were investigated by the agarose gel electrophoresis method. Results showed that both cycloartelastoxanthone (9) and artelastoheterol (11) possessed the protective effects against DNA damage caused by•O . Compound 9 showed potent inhibitory effect on DNA scission induced by •O .
