類風濕性關節炎是慢性發炎性疾病,其發病機制是複雜的,多因素,包括遺傳,免疫失調,和環境的影響,其詳細致病機轉至今仍不明。我們研究的目的在探討Gadd45b基因表現及基因多型性與類風濕性關節炎的相關性。 實驗選取90位正常人,127位RA病人,抽取周邊血液單核淋巴細胞 (PBMCs)的total RNA,與萃取buffty coat細胞的Genomic DNA,利用real-time PCR方法偵測Gadd45b基因表現。此外,選取206位正常人,261位RA病人以ABI公司的TaqMan基因型鑑定進行基因多型性研究。統計分析Gadd45b基因表現及單一核苷酸基因多型性(SNP)與臨床徵狀之間的關係。 結果顯示,RA病人PBMCs的Gadd45b基因表現量較正常人低(p=0.02),與臨床徵狀類風濕性結節及RA疾病嚴重性有相關性,所挑選的三個SNP(Gadd45b-438A/C、-712C/T、+459 C/G)與類風濕性關節炎沒有相關性。 我們的實驗顯示,RA病人之PBMCs其Gadd45b基因表現量較正常人低,且與臨床徵狀類風濕性結節及RA疾病嚴重性有相關性,但與所挑選的三個SNP及臨床症狀沒有相關性。這些研究將有助於進一步了解RA的致病機轉和治療上一個新的思考方向。
Rheumatoid arthritis(RA) is a chronic inflammation disease, its pathogenesis is complex, multifactorial, including genetic, immune disorders, and environmental impacts, the detailed pathogenesis is still unknown. Our research aims to investigate Gadd45b gene expression and gene polymorphism with rheumatoid arthritis correlation. Ninety healthy contols,and 127 patients with RA were enrolled in this study.Total RNA and Genomic DNA of buffy coat cells were extract from peripheral blood mononuclear cells (PBMCs). The Gadd45b gene expression was measured by the method of real-time PCR. In addition, 206 healthy controls and 261 patients with RA were enrolled for Gadd45b polymorphism study. Gadd45b single-nucleotide polymorphism (SNP) research was manipulated according to ABI (Applied biosystems incorated) TaqMan genotyping assays. Statistical analysis Gadd45b gene expression and single nucleotide polymorphism (SNP) and the relationship between clinical symptoms. This study demonstrated that Gadd45b gene expression in PBMCs was significantly lower in patients with RA than in controls (p=0.002). This study also showed that Gadd45b gene expression in PBMCs were associated with the clinical manifestations of RA. The single nucleotide polymorphisms were not associated with the RA. Our data shows that Gadd45b gene expression in PBMCs was significantly lower in patients with RA than in controls. Gadd45b gene expression in PBMCs were associated with the clinical manifestations of RA. The single nucleotide polymorphisms were not associated with the RA.The result may offer new etiological and therapeutic insight into RA.
