探討 Brefeldin A 對於肺癌細胞毒殺之影響

摘要


目的:本研究評估Brefeldin A 在肺癌細胞株A549、H1299的細胞毒性以及其抑制肺癌幹細胞 (Lung Cancer Stem Cell) 的潛能。 實驗設計:首先在一開始採用MTT 與WST-1聯合測試Brefeldin A對於兩肺癌細胞株A549及H1299進行毒殺後的存活率且同時也測試對於LCSC的影響,而後採用體外傷口修復試驗測試其抑制細胞移行的能力,接著進一步地使用洋菜膠及一般性培養多孔盤測試其對於癌症幹細胞特性的影響。 結果:Brefeldin A可殺除肺癌幹細胞,然而卻不對一般貼附培養的A549及H1299肺癌細胞造成破壞,在細胞移行能力試驗當中,明顯地可使細胞刮痕較為緩慢地修復,除此之外,對於使用洋菜膠培養之懸浮細胞,不管是在尺寸大小或是數量皆有明顯地影響,另外也可從一般性多孔盤培養看出其可顯著地影響其由單獨細胞形成細胞群落能力影響。 以上結果給予癌症治療一個新的方向,BFA在上述實驗中可有效地抑制肺癌幹細胞生長且可防止細胞移行,若是要針對癌症發展目標治療的新藥物。可針對上述實驗結果做進一步地探討。

Purpose: To investigate the cytotoxic effect of Brefeldin A in two different lung cancer cell lines, A549 and H1299, and the potential anti-cancer effect on target LCSC (lung cancer stem cell). Experimental design: First, we evaluated the cytotoxic effect of Brefeldin A on cell viabilities of lung cancer cell lines A549 and H1299 as well as LCSC, and then we investigated the inhibitory effect on cancer cell migration in vitro by a wound healing assay. Moreover, a colony formation assay using soft agar in multi-well plates was performed to measure the effect of Brefeldin A on the stemness of cancer cells. Results: Brefeldin A was able to specifically kill LCSCs. There was no effect on A549 and H1299 in attachment culture. In the cell migration experiments, the tumor cells exhibited significantly slower wound closure after Brefeldin A treatment. Moreover, significant reductions in colony size and number were observed in colony formation assay in both cell lines after Brefeldin A treatment. The above results provided a new heading in cancer treatment where Brefeldin A effectively inhibited lung cancer stem cell growth and prevented metastasis in lung cancer cell which can be further investigated in the future to develop new therapy in cancer treatment.
