摘要


本論文的研究主題是利用大體積樣品堆積(large volume sample stacking; LVSS)之毛細管電泳法對人體血漿和腹腔液中抗生素藥物piperacillin,建立一個簡單、選擇性及敏感性高之定量分析法。分離之背景電解質(background electrolyte; BGE)為: 60 mM 磷酸鈉鹽緩衝溶液(pH 6),內含0.005% polyethyleneoxide (PEO)是用來抑制毛細管內壁電滲透流;使用液相液相萃取法來處理血漿和腹腔液檢品。並以cephalothin作為內部標準品。樣品以0.5 psi壓力注入70s到毛細管中,再應用電壓方式(-15 kV)進行分離,且偵測波長為200 nm。對影響分離的因素: 磷酸鈉鹽緩衝溶液的pH值和濃度、PEO濃度。線性範圍為0.5-10.0 μg/mL的piperacillin濃度。此分析方法可應用於IV給藥後病人血漿和腹腔液中piperacillin之含量分析。

A sensititive high performance capillary zone electrophoresis (CZE) combining on-column large volume sample stacking (LVSS) injection has been developed for determination of piperacillin in human plasma and peritoneal fluid. A sample pretreatment by means of liquid-liquid extraction (dichloromethane) with subsequent quantitation by LVSS-CZE was used. The separation of piperacillin was performed using a background electrolyte containing 60 mM phosphate buffer (pH 6) with 0.005% polyethyleneoxide (PEO) as a dynamic coating capillary wall to inhibit electroosmotic flow (EOF). The sample was hydrodynamic injection (0.5 psi, 70s) to introduce sample and the applied voltage was -15 kV and on-column detection at 200 nm. Several parameters affecting the separation and sensitivity of piperacillin were studied, including the pH and concentration of phosphate buffer and concentration of PEO. The linear range of the method for determination of the test drug, in plasma and in peritoneal fluid using cephalothin as an internal standard (IS), was all over the range 0.5-10.0 μg/mL for piperacillin. The application of the proposed method for determination the concentration profiles of piperacillin in plasma and in peritoneal fluid collected after IV injection tazocin for severe intra-abdominal infection patient was demonstrated.
